Rats with physically disconnected hypothalamo‐pituitary tracts no longer contain vasopressin‐oxytocin gene transcripts in the posterior pituitary lobe

Abstract: In rats, vasopressin-and oxytocin-encoding mRNAs are present in the posterior but absent in the anterior lobe of the pituitary gland. RNase protection experiments indicate that in the posterior pituitary and hypothalamus identical transcriptional start points are used. Furthermore, the two transcripts from posterior pituitary and hypothalamus show identical nucleotide sequences. Animals operated by paired electrical lesions in such a way that connections between the supraoptic nucleus (SON) and paraventricular… Show more

“…In the chorio-decidua, this difference is not likely due to differences in the coding region of the mRNA since both the 5'-and 3'-specific probes were completely protected by RNA from these tissues. The difference in size may be due to a variation in the poly(A) tail length, as has previously been noted in bovine hypothalamic and ovarian OT mRNA (30 Our results also demonstrate that OTmRNA the human pituitary and corpus luteum thus suy vious studies in mammals (35)(36)(37)(38) (39).…”

“…However, biochemical and morphological studies have shown that nerve cells that constitute the rat hypothalamo-neurohypophyseal tract harbor various RNA species-for instance, arginine vasopressin (AVP) and oxytocin (OT) mRNAs (9)(10)(11) as well as BC1 RNA (12)-in the axonal domain. In the homozygous Brattleboro rat (BB rat), mutant AVP transcripts are not detectable in the axonal compartment (13) even though the cell bodies of magnocellular neurons contain this mRNA (14).…”

“…Interestingly, this is accompanied by an elongation of their poly(A) tails although the physiological significance remains to be determined [4][5][6]. Both AVP and OT mRNAs are subject to axonal targeting [7][8][9] and hyperosmolar stimuli give rise to a differential increase in the level of AVP (17-fold) and OT (3-fold) mRNAs in the axonal compartment [8,[10][11][12]. The axonal transcripts exhibit shorter poly(A) tracts than their counterparts in the cell bodies [2], and a poly(A) tail polymorphism following osmotic challenge is not observed for these transcripts [12].…”
