Rats with a Glucocorticoid‐Induced Catabolic State Show Symptoms of Oxidative Stress and Spleen Atrophy: The Effects of Age and Recovery

Abstract: In this study we wanted to determine whether changes in antioxidant profile could follow the catabolic effects of glucocorticoids. We also wanted to compare resistance to glucocorticoid overload in young and old rats. To address these questions, whole body catabolism was induced by the administration of dexamethasone (Dex) at either 2 mg/kg bodyweight/day to young (6 weeks old) or 0.5 mg/kg body-weight/day to old (94 weeks old) rats. Bodyweight loss of pair-fed rats not given Dex was only 2% in the young rats … Show more

“…At the study end point, prednisolonetreated mice in the mdx mouse trial showed significant losses of both body weight and spleen weight (Fig. 4, B and C), which is consistent with previous reports of deleterious side effects in rats (Orzechowski et al, 2002), whereas anecortave at higher doses showed none of these side effects. Likewise, in the dysferlin-deficient (SJL) trial, prednisolone, but not anecortave, induced a significant loss of spleen weight [control, 6.09 Ϯ 0.34 g; prednisolone, 4.11 Ϯ 0.23 g ‫,ء(‬ p Ͻ 0.05); anecortave, 6.52 Ϯ 0.59 g].…”

“…Glucocorticoid administration increases plasma markers of oxidative stress in mammals (Orzechowski et al, 2002;Lin et al, 2004). It is unclear how these stress hormones might affect telomerase and telomeres in human PBMCs.…”

“…The exact mechanism of action of prednisolone in NS has not been elucidated yet, although it is believed to be caused by immunosuppressive and antiinflammatory properties (3). Moreover, the results of some experimental studies suggest that steroids directly or indirectly impair the antioxidant reactions and lead to overproduction of ROS (46,47). However, the study of Kawamura et al (48) suggested that glucocorticoids may have a therapeutic role through the stimulation/augmentation of the activity of antioxidant enzymes.…”
