Rational strain improvement for surfactin production: enhancing the yield and generating novel structures

Abstract: Surfactin, one of the most powerful microbial surfactants, is a lipopeptide-type biosurfactant which combines interesting physicochemical properties and biological activities. However, the high cost caused by its low productivity largely limits the commercial application of surfactin. Hence, many engineered bacterium have also been used to enhance surfactin biosynthesis. This review briefly summarizes the mechanism of surfactin biosynthesis, highlighting the synthesis pathway of N-terminally attached fatty aci… Show more

“…ORF f denotes for the enzyme acyl-CoA dehydrogenase (ACAD) which catalyses the β -oxidation of branched-chain and unsaturated fatty acids [ 63 ]. The dehydrogenase complex may also take part in the conversion of the CoA precursors to the respective fatty acid chains [ 64 ]. ORFs e and d code for the MaoC family dehydratase and CoA ester lyase, respectively.…”

“…We hypothesize that the reduction of surfactin production might be due to altered gene regulation by a mutation in the comA gene. However, it is unclear to what extend the ComA protein level is affected due to mutation in the translation initiating methionine and whether the potentially altered level of this transcription factor in strain MB9_B4 mitigates sufficient binding to the promoter region of srf to activate its transcription (50). Repair of the comA mutation could potentially corroborate if this single mutation is causing the reduced surfactin production in MB9_B4.…”

“…The implications of overwintering and the resting stages of pathogens could also be worked out to reduce the inoculum loads from croplands. The microbial strain may be engineered for the novel structure of surfactin production [206]. Further, advancement in the bioinformatics assisted molecular and dynamics simulation studies can also help to decipher the molecular and biochemical mechanisms.…”
