In a recent one-year feeding study, we observed no adverse effects on tissue level in organs of rats fed with the genetically-modified maize MON810. Here, we assessed RNA expression levels of 86 key genes of the apoptosis-, NF-кB-, DNA-damage response (DDR)-, and unfolded-protein response (UPR) pathways by RT-qPCR in the rat liver. Male and female rats were fed either with 33% MON810 (GMO), isogenic- (ISO), or conventional maize (CONV) and RNAs were quantified from eight rats from each of the six feeding groups. Only Birc2 transcript showed a significant (p ≤ 0.05) consistent difference of ≥1.5-fold between the GMO and ISO groups in both sexes. Unsupervised cluster analysis showed a strong separation of male and female rats, but no clustering of the feeding groups. Individual analysis of the pathways did not show any clustering of the male or female feeding groups either, though transcript levels of UPR pathway-associated genes caused some clustering of the male GMO and CONV feeding group samples. These differences were not seen between the GMO and ISO control or within the female cohort. Our data therefore does not support an adverse effect on rat liver RNA expression through the long-term feeding of MON810 compared to isogenic control maize.

Background: Roundup Ready became the most cultivated genetically modified (GM) crop in the world. The aim of this study was to evaluate the effects of a diet containing GM soybean meal (RUR) on male and female Wistar rats. The body weights, food consumption, hematology, serum biochemistry, organ weight, and histopathological examination were compared between rats fed on GM soybean and those fed non-GM soybean after consumption of test diets for 18 weeks. Methods: Sixty weaned Wistar albino rats (30 females and 30 males) were randomly assigned to three groups of ten rats for each group per each sex. All rats have been fed either a diet containing 0% GM soybean meal (control), 50% GM soybean meal (T1), and 100% GM soybean (T2). The body weights, food consumption, hematology, serum biochemistry, organ weight, and histopathological examination were compared between rats fed GM soybean and those fed non-GM Soybean after consumption of test diets for 18 weeks. Results: Examined hematological parameters did not indicate a statistically significant difference between the GM soybean-fed groups (T1 and T2) and their respective controls, though the WBCs of the male of treated groups (T1 and T2), platelets count, and hemoglobin (HB) of the female of (T1) group were slightly elevated. The examined biochemical blood parameters showed that the level of triglycerides (TG) was statistically significantly higher in the female treated groups (T1 and T2) and lactate dehydrogenase (LDH) was significantly higher in treated groups compared to their respective controls. Elevated levels of alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and creatinine were observed in some of the treated groups. Histopathological examination of selected organs (liver, kidney, and pancreas) and testes or ovaries and female mammary gland revealed histological alterations among different treated groups (T1 and T2) with their respective controls. Conclusion: The obtained results indicate the necessity to revise the safety standards of consumption of diets containing GM soybean.

Regulatory-compliant rodent subchronic feeding studies are compulsory regardless of a hypothesis to test, according to recent EU legislation for the safety assessment of whole food/feed produced from genetically modified (GM) crops containing a single genetic transformation event (European Union Commission Implementing Regulation No. 503/2013). The Implementing Regulation refers to guidelines set forth by the European Food Safety Authority (EFSA) for the design, conduct, and analysis of rodent subchronic feeding studies. The set of EFSA recommendations was rigorously applied to a 90-day feeding study in Sprague-Dawley rats. After study completion, the appropriateness and applicability of these recommendations were assessed using a battery of statistical analysis approaches including both retrospective and prospective statistical power analyses as well as variance-covariance decomposition. In the interest of animal welfare considerations, alternative experimental designs were investigated and evaluated in the context of informing the health risk assessment of food/feed from GM crops.
