Rapid propagation of agave by in vitro tissue culture

Abstract: A procedure for rapid propagation of Agave (A. eantala Roxb., A. fourcroydes Lem. and A. sisalana Perrine, (Agavaceae) have been developed. The explants were excised from stolon plantlets, sterilized and cultivated on Murashige and Skoog (MS) basal medium containing 2% sucrose, 10% coconut water and 0.8% agar. The addition of following combination of growth substances --0.075 mg l 1 naphthalenacetic acid (NAA) + 0.1 mg 1-~ indolylbutyric acid (IBA) + 0.5 mg 1-l kinetin (KIN) caused an extensive proliferat… Show more

“…These hormones have been used for the promotion of growth in several plants that form stolons [18,19]. We studied 64 culture conditions with various combinations of kinetin (0, 0.1, 0.5, 2.5 mg/L), GA3 (0, 0.01, 0.1, 1 mg/L), and NAA (0, 0.01, 0.1, 1 mg/L) for 4 weeks and measured the number of newly formed ramets and the weights of After growing aseptic plants on solid medium for more than one month, the plants were transferred to liquid medium in a plastic container.…”

“…A straw was placed over the ovary to prevent oviposition by moths. Ovules were removed from the ovary as described in the tetrazolium experiment, and were placed on media made from a modified recipe of Murashige basal media agar (Murashige 1974;Binh et al 1990). The modified recipe contained 8% sucrose, 4.3 g Murashige and Skoog salts, 0.4 mg thiamine, 10 mg inositol, 2.0 mg IAA, and 1% agar per liter of media, and the pH was adjusted to 6.0.…”
