Rapid colorimetric determination of free fatty acids

Abstract: In 1964, a method was described for the determination of free fatty acids (FFAs) in vegetable oil. This paper describes an expansion of that work, improving the sensitivity and reproducibility of the method, as well as examination of solubilities of the copper soaps as a function of chain length and unsaturation. Involvement of the micellar structure was reviewed. Finally, a procedure is described that permits very rapid determination of FFA at the 2.0-14.0/~mol (0.5-4.0 mg oleic acid) level, and the results w… Show more

“…Reducing sugars content was determined by following the method of Nelson (1944). The content of phospholipids was estimated by determining phosphorus content as described by Ames (1966), glycolipids by measuring glucose content according to Roughan and Batt (1968), free fatty acids by copper complex formation as described by Lowry and Tinsley (1976) and sterols by using ferric chloride according to Zak (1957), all spectrophotometrically. The content of total glycerides was calculated by the difference of total lipid classes from 100.…”

“…After removal of excess solvent, the fat was dried, weighed and analysed for FFA by colorimetric procedure (Lowry and Tinsley 1976). The amount of FFA was calculated as palmitic acid equivalent from the calibration curve using different aliquots of standard palmitic acid and expressed as% on lipid basis.…”
