Radioimmunoassay, complement fixation and counter-immunoelectrophoresis in the laboratory diagnosis of hepatitis B

Abstract: Three hundred and fifty specimens from a selected group of hospital patients were tested for the presence of hepatitis B antigen (HB-Ag) by radioimmunoassay (RIA), micro-complement fixation test (CFT) and eounter-immunoelectrophoresis (CIEP).I~IA detected 82 (23.7 per cent), CFT 63 (18 per cent), and CIEP only 43 (12.2 per cent) HB-Ag positive sera. All sera found positive by CFT and CIEP were also positive by I~IA. No serum was found positive by CFT or CIEP that was negative by RIA.The problem of anticomplemen… Show more

“…However, it lacks a certain degree of specificity by showing false positive results. This fault could be corrected satisfactorily by absorbing the sera with anti-HB-Ab and retesting (7). Modification of the Ausria-I failed to solve the problem of specificity satisfactorily in our hands (9).…”

“…A variety of tests are generally available for the laboratory diagnosis of hepatitis B antigen (HB-Ag), such as the agar gel diffusion, counterimmunoelectrophoresis (CIEP), complement fixation test (CFT), immunoelectromicroscopy, reversed passive hemagglutination (RPH), and direct radioimmunoassays (RIA) (3)(4)(5)(6)(7).…”

“…Der Ausria 11 125-Test wurde in der üblichen Prozedur mit Inkubationszeiten von zwei Stunden und einer Stunde im Wasserbad bei 45 oe und Zählung der Impulse für eine Minute ausgeführt. Zur Verfügung standen Kits der Lot-Nummer 71.959.BX 1. 4.…”

“…CEP, although a quick test, is far less sensitive than RIA. Various other tests have been described, such as Ouchterlony double diffusion, red cell agglutination, reverse passive hemagglutination, latex agglutination, complement fixation, and immunoelectron microscopy (2,4,5,10,11), to name a few. Recently, a rapid hemagglutination test has been described (Wellcome Laboratories, England) which uses turkey erythrocytes coated with horse antibody to hepatitis B antigen purified by affinity column chromatography.…”
