RAB11-Mediated Trafficking and Human Cancers: An Updated Review

Abstract: Many disorders block and subvert basic cellular processes in order to boost their progression. One protein family that is prone to be altered in human cancers is the small GTPase RAB11 family, the master regulator of vesicular trafficking. RAB11 isoforms function as membrane organizers connecting the transport of cargoes towards the plasma membrane with the assembly of autophagic precursors and the generation of cellular protrusions. These processes dramatically impact normal cell physiology and their alterati… Show more

“…Whether L. monocytogenes interferes with similar mechanisms is currently unclear, and future work will be needed to characterize the interplay between mitochondrial energy metabolism, endocytic recycling, and infection. In particular, investigations into the contributions of the three Rab11 isoforms are of interest, as the view emerges that they have distinct functions in endocytic recycling ( Ferro et al., 2021 ).…”

“…Two major mechanisms of ILV formation exist in animals: Endosomal Sorting Complex Required for Transport (ESCRT)-mediated pathway and ceramide-mediated pathway ( van Niel et al, 2018 ; Mathieu et al, 2019 ). Mechanisms that drive the mobilization of secretory MVBs and fusion with the plasma membrane require the participation of Rab family proteins (Rab11, Rab35, and Rab27), and SNARE family proteins (vesicle-associated membrane protein 7 and YKT6 V-SNARE homolog; Hsu et al, 2010 ; Ostrowski et al, 2010 ; Kowal et al, 2014 ; Tian et al, 2020 ; Ferro et al, 2021 ). In fact, in animal cells, MVBs also fuse with autophagosomes and further form amphisomes following the release of EVs containing autophagy components ( Klionsky et al, 2014 ; Jeppesen et al, 2019 ).…”

“…Interestingly, overexpression of Rab11 by plasmid transfection significantly increased LDLR expression (Figure S2B). Existing reports suggest that Rab11 plays a housekeeping role in regulating intracellular pathways between endocytic recycling, lysosomal degradation and exocytosis 24 . Moreover, inhibition of Rab11‐ mediated reverse transport of cellular contents can promote the activation of its degradation through lysosome 25 .…”
