Quorum Sensing Promotes Phage Infection in Pseudomonas aeruginosa PAO1

Abstract: Phage resistance is a major limitation of phage therapy, and understanding the mechanisms by which bacteria block phage infection is critical for the successful application of phage therapy. In the present study, we found that Pseudomonas aeruginosa PAO1 uses las QS to promote phage infection by upregulating the expression of galU , which is necessary for the synthesis of phage receptor lipopolysaccharides.

“…Moreover, alternate dark/light led to alternate disassembly and assembly of the bacterial cells, and N-butanoyl-l-homoserine lactone (C4-HSL). 50,51 The neighboring cells sense these molecules and respond to them by the LasR/3-oxo-C12-HSL or RhlR/C4-HSL complex, further controlling the expression of genes involved in diverse bacterial behaviors, such as biofilm formation, production of secondary metabolites, and swarming motility. 52−54 Because of the presence of the magnetic core and the MSNs, the Mag@ MSN-SP+ChiPSP assemblies may load the small-molecularweight QSM (e.g., 3-oxo-C12-HSL, Scheme 1) and release them by AMF heating-enhanced molecule diffusion (Figure 5a).…”

“…To measure the loading and release capacity of the assemblies, an QSM-reporting E. coli strain (i.e., EcplasGFP) was constructed by transformation of E. coli BL21 with the QSreporting plasmid PplasGFP, in which the expression of GFP was controlled by the QS-responsive promoter plas to reflect QS levels (Figures 5a and S12). 50,51 With the increase in QSM concentration from 5 to 100 μM, EcplasGFP showed an increased GFP intensity (Figure S13a). Especially, there was a linear relationship between the QS concentrations from 5 to 30 μM and GFP intensity (Figure S13b).…”

“…Quorum sensing plays an important regulatory role in biofilm formation, bacterial persistence, and so on [ 10 ]. For instance, Xuan et al found that QS upregulated the expression of lipopolysaccharide synthesis and promoted biofilm development in P. aeruginosa [ 11 ]. Tsao et al re-engineered the native QS regulon, in which Escherichia coli secreted QS signal autoinducer-2 (AI-2) initiated and guided the high-level expression of recombinant proteins [ 12 ].…”
