Quantitative Structure-Activity Relationships of Chemicals Acting by Non-polar Narcosis—Theoretical Considerations

Abstract: Theoretical quantitative structure-activity relationships have been established based on narcotic mechanisms of action and toxicity data to the fathead minnow, Daphnia magna and Vibrio ®scheri. The results con®rmed that hydrophobicity is important in modelling narcosis. However, the results of the present investigation also con®rmed that molecular bulk may play an important role in toxicity to aqueous organisms, especially in the toxicity to organisms with lower lipid content, for example, V. ®scheri. The resu… Show more

“…toxicity is increasing linearly with lipophilicity. 5,[22][23][24]43 Consideration of the QSARs developed in this study shows an improvement in the models when utilising CS-weighted regression. The improvement is both the statistical fit but also the slope for log K OW which approaches one when employing CS-weighting, i.e.…”

“…The concentration values causing a 50% inhibition of bioluminescence after 15 or 30 min exposure to V. fischeri (expressed as IBC 50 ) for 1239 compounds were taken mainly from several references (Kaiser and Palabrica, 1991;Cronin et al, 2000;Zhao et al, 1998;Terasaki et al, 2009;Dearden et al, 2000;Aruoja et al, 2011;Jones et al, 2011;Qin et al, 2010;Shi et al, 2012;Villa et al, 2012). No significant difference was observed in the two toxicity endpoints.…”

“…found that the hydrophobicity also correlated well with polar narcosis to the fathead minnow for 39 phenols and anilines. Many investigators have used the octanol/water partition coefficient dependent QSAR for predicting the toxicity of ARTICLES ENVIRONMENTAL CHEMISTRY non-polar and polar narcotic chemicals [2,6,7,8] . It is also called the baseline toxicity model for chemicals acting by non-polar narcosis.…”

“…Poor correlations were found between logP and toxicity to different kinds of aquatic organisms for these reactive chemicals. These compounds had to be removed from the data sets to obtain the prediction models between logP and toxicity for non-polar and polar chemicals [7,12,14] . The toxicity process involves both toxicant transport to the site of interaction on a biomacromolecule and interaction between the chemical at the active site.…”
