Quantitative real-time PCR detection and analysis of a lumpy skin disease outbreak in Inner Mongolia Autonomous Region, China

Abstract: Lumpy skin disease (LSD) is a severe disease of bovine characterized by nodules on the skin, mucous membranes, and profuse nasal discharge which causes severe economic losses. In October 2020, an LSD outbreak case was found in Inner Mongolia Autonomous Region, China. A total of 1,206 cattle were sold from the same imported animal quarantine field to 36 farms after the quarantine period finished, and over 30 farmers reported symptoms such as skin scabs found in newly arrived cattle shortly after that. A large-s… Show more

“…The con rmed cases of LSDV revealed 14.28% of positivity in blood, 95.04% of positivity in skin scab and 66.66% of positivity in nasal swab by speci c conventional PCR. Similarly the higher percentage of scab samples were found positive for LSDV in this study are in agreement with result of recent reports [40,41,42,43,44,27]. Moreover, skin nodule/scab samples are the most preferred sample types for LSD diagnosis and LSDV genome can be detected in skin lesions up to 92 days after infection [40].…”

“…In outbreaks these viruses display variability in morbidity, mortality and severity of clinical disease. The patterns of virus replication and secretion between experimental infection of classical LSDV (Babiuk et al, 2008b) and recombinant LSDV observed in the field are similar (Li et al, 2022).…”

“…In recent years, Taqman-Based qPCR methods have been developed, and widely applied for detecting LSDV [20][21][22], since Taqman-Based qPCR method showed higher sensitivity than the SYBR Green qPCR methods and PCR methods [23]. In this study, a Taqman-Based qPCR approach was successfully established for LSDV detection, the ampli cation e ciency (E) and correlation coe cient (R 2 ) of the standard curve linearity was 0.9968 and 98.48%, respectively.…”
