Quantitative Collection and Enzymatic Activity of Glucose Oxidase Nanotubes Fabricated by Templated Layer-by-Layer Assembly

Abstract: We report on the fabrication of enzyme nanotubes in nanoporous polycarbonate membranes via the layer-by-layer (LbL) alternate assembly of polyethylenimine (PEI) and glucose oxidase (GOX), followed by dissolution of the sacrificial template in CH2Cl2, collection, and final dispersion in water. An adjuvant-assisted filtration methodology is exploited to extract quantitatively the nanotubes without loss of activity and morphology. Different water-soluble CH2Cl2-insoluble adjuvants are tested for maximal enzyme ac… Show more

“…Moreover, it is a versatile technique which may be applied to almost all substrates independently of their shape, chemical composition and physical properties, including nanoporous materials. 16 Bioactive nanotubes/nanowires have been obtained by some of us [17][18][19] and other groups. [20][21][22] They were successfully used for different purposes, such as the control of preosteoblast adhesion.…”

“…In addition, the enzymatic activity of (bPEI/GOX) 6 collected by the adjuvant-assisted method is preserved, as will be extensively reported elsewhere. 57 As observed in Figure 7a−e, LbL-assembled nanotubes have a tendency to bend, fold, and form loops after being dispersed in aqueous solution. In other words, there is a difference in nanotubes rigidity prior and after being dispersed in aqueous solution.…”

“…There have been other reports on the preparation of LbL films by electrostatic interaction using QCM [36]. LbL films using DNA or GOx driven by electrostatic interactions have been reported by Zhang et al [37,38] and Kakade et al It is considered that DNA and GOx are both present in the negatively charged layers because DNA and GOx adsorb to positively charged polymers. Figure 4 shows an AFM image and a depth profile of a dried (H-PEI/DNA + GOx) 5 film.…”
