Quantitative Bioimaging of Platinum via Online Isotope Dilution-Laser Ablation-Inductively Coupled Plasma Mass Spectrometry

Abstract: A new calibration strategy for elemental bioimaging based on online isotope dilution analysis (IDA) and laser ablation-inductively coupled plasma mass spectrometry (LA-ICPMS) was developed and applied for the quantification of platinum in rat kidney tissues. A dry Pt spike aerosol was added in a post-cell setup, and the naturalPt/Pt isotope ratio of the sample aerosol from laser ablation was changed accordingly. Spike mass flow determination was carried out based on reversed IDA using a reference standard. Qua… Show more

“…Fernandez et al determined the mass fraction by parallel analysis of a CRM with a known concentration 161 and applied the method to the quantification of powdered samples. Using a customized reference material, Bauer et al 162 followed a similar strategy for the spatially resolved quantification of Pt in biological tissue. Pickhardt et al 163 mounted a microflow nebulizer in the ablation cell which mixed the dry sample aerosol with a wet isotopically enriched standard.…”

“…[2][3][4] Different research and industrial 5 fields are interested in this technique and the areas of application are wide spread. For example, the LA-ICP-MS is used in geology 6 , medicine 7,8 , but also for the provenance analysis 9 or forensic science. 10 Although numerous applications of LA-ICP-MS have been carried out during the last years, it is still challenging to make them quantitative and traceable to the SI (Système international d'unités).…”

“…LA-ICP-MS can provide quantitative results as shown by previous studies in which either matrix-matched external calibration, internal calibration with suitable solutions, or isotope dilution analysis was used. [18][19][20][21] Further prove of this concept came from complementary ICP-MS analyses of standard concentrations aer microwave digestion. 22,23 Since LA-ICP-MS may also be used to detect metal-labelled biological targets [24][25][26] we combined the quantitative mass spectrometric detection of silver with a gold and uorescence-based immunohistochemical detection technique for biological target structures.…”
