Several commercial beers have been analyzed for their content of β-glucans, pentosans and their degradation products using high performance liquid chromatography, thin layer chromatography and chemical and enzymic analysis procedures. The beers tested contained high levels of residual high molecular weight pentosan, but much less high molecular weight β-glucan. The beers also contained sizeable levels of oligosaccharides, especially trisaccharides, reflecting the incomplete degradation of polymeric materials in malting and mashing and the inability of yeast to ferment them. There is substantially more β-linked glucosyl material in beer than pentosyl substances, although the higher molecular weight of the latter probably makes it more likely to represent soluble fibre. In respect of fibre claims for the beers examined, even for the beer containing the least pentosan, it seems that less than a litre of the product would afford sufficient material.

Quantification of Wheat TAXI and XIP Type Xylanase Inhibitors: A Comparison of Analytical Techniques

To date, three different techniques are available for the quantification of TAXI and XIP type proteinaceous xylanase inhibitors in cereals. A first approach is based on the determination of the residual activities of xylanases (also referred to as endo‐1,4‐β‐d‐xylanases, EC 3.2.1.8), which are specifically inhibited by these inhibitors, after incubation with sample containing the inhibitors. The other two techniques are immunoblotting and ELISA which are based on recognition of TAXI and XIP proteins by specific antibodies. TAXI, as well as XIP, are easily extracted by aqueous buffers. Hence, the large difference in their concentrations (2–10 fold higher for XIP than for TAXI in whole meal) is not caused by differences in extractability. The repeatabilities of the three techniques are comparable. The intra‐assay and inter‐assay coefficients of variation were 6–7 and 10–14%, respectively, which is in the range of values described for methods to quantify other compounds in plant and animal tissues. The three methods give comparable results, suggesting they have similar accuracies. The choice of the technique to be used will depend not only on the sensitivity and dynamic range needed, but also on its technical simplicity and the need for high‐throughput analysis.

Proteinaceous inhibitors of microbial xylanases

At the end of 1990s two structurally different proteinaceous inhibitors of xylanases were discovered in the grain of wheat (Triticum aestivum). They were named TAXI (T. aestivum xylanase inhibitor) and XIP (xylanase-inhibiting protein). Later it was shown that TAXI and XIP in wheat are present in several isoforms encoded by different genes. TAXI- and XIP-like inhibitors have also been found in other cereals-barley, rye, rice, maize, etc. All these proteins can specifically inhibit activity of fungal and bacterial xylanases belonging to families 10 and 11 of glycoside hydrolases, but they do not affect endogenous enzymes produced by plants. A common viewpoint is that the presence of proteinaceous inhibitors in cereals is a response of plants to pathogenic attack by microorganisms. A few years ago, an inhibitor of a third type was discovered in wheat. It was named TLXI (thaumatin-like xylanase inhibitor) because of its similarity to the thaumatin family of plant proteins. In this review, the occurrence of proteinaceous inhibitors of xylanases in different cereals, their specificity towards fungal and bacterial enzymes, as well as structural features responsible for enzyme sensitivity to various types of inhibitors are discussed.
