The rapid, highly-sensitive and ecologically greener reversed-phase (RP)/normal-phase (NP) high-performance thin-layer chromatography (HPTLC) densitometric technique has been developed and validated for the determination of trans-resveratrol (TRV). The reversed-phase HPTLC-based analysis of TRV was performed using ethanol–water (65:35, v v−1) combination as the greener mobile phase, while, the normal-phase HPTLC-based estimation of TRV was performed using chloroform–methanol (85:15, v v−1) combination as the routine mobile phase. The TRV detection was carried out at 302 nm for RP/NP densitometric assay. The linearity was recorded as 10–1200 and 30–400 ng band−1 for RP and NP HPTLC techniques, respectively. The RP densitometric assay was observed as highly-sensitive, accurate, precise and robust for TRV detection in comparison with the NP densitometric assay. The contents of TRV in commercial formulation were recorded as 101.21% utilizing the RP densitometric assay, while, the contents of TRV in commercial formulation were found to be 91.64% utilizing the NP densitometric assay. The greener profile of RP/NP technique was obtained using the analytical GREEnness (AGREE) approach. The AGREE scales for RP and NP densitometric assays were estimated 0.75 and 0.48, respectively. The recorded AGREE scale for the RP densitometric assay indicated that this technique was highly green/the ecologically greener compared to the NP densitometric assay. After successful optimization of analytical conditions, validation parameters, AGREE scale and chromatography performance, the RP densitometric assay with univariate calibration was found to be better than the NP densitometric assay for the analysis of TRV.

Metabolomic Profiling and Identification of Antioxidant and Antidiabetic Compounds from Leaves of Different Varieties of Morus alba Linn Grown in Kashmir

Mulberry ( Morus alba L. ) is commonly cultivated in Asian countries as a traditional medicine and food supplement. Four Kashmiri Morus alba varieties ( Zagtul , Chtattatual , Chattatual Zaingir , and Brentul Kashmir ) were evaluated for their proximate composition, mineral content, total phenolic and flavonoid content, antioxidant potential, and antihyperglycemic activity. Furthermore, TLC-MS-bioautography was used for the identification of antioxidant and antidiabetic compounds in the best active extract. Lastly, UPLC-MS was employed for metabolomic profiling of the best variety of M. alba . Among all the varieties, the Zagtul variety was found to have the highest phenolic (71.10 ± 0.44 mg GAE/g DW) and flavonoid (53.22 ± 0.69 mg rutin/g DW) content. The highest antioxidant potential (DPPH) with an IC 50 value of 107.88 ± 3.8 μg/mL was recorded for the Zagtul variety. Similarly, α-amylase and α-glucosidase inhibition for antidiabetic potential with IC 50 74.76 ± 6.76 and 109.19 ± 5.78 μg/mL, respectively, was recorded in Zagtul variety. TLC-MS-bioautography for identification of bioactive compounds revealed the presence of chlorogenic acid for antioxidant potential and 1-deoxynojirimycin (DNJ) and syringic acid for antidiabetic potential. Further, bioactive compounds responsible for diverse functions of M. alba were confirmed by UPLC-MS in both negative and positive modes. However, major compounds in the Zagtul variety were identified as chlorogenic acid, moracin N, gallic acid, ferulic acid, morin, 1-deoxynojirimycin, and syringic acid. Hence, based on our findings, it can be concluded that M. alba leaves can be consumed as a promising dietary supplement and can be formulated as phytopharmaceutical for the management of various metabolic disorders.

Introduction The quantitative analysis of trace resveratrol and polydatin in plant tissues is suitable for elucidation of the compounds' mechanisms of action. Objectives The main objective of this work was to develop a feasible and effective sample pretreatment method to measure the concentrations of resveratrol and polydatin in complex samples. Methodology A polymer sorbent, poly(2‐mercaptobenzimidazole), was electrochemically prepared and utilized for selective extraction, while resveratrol and polydatin were used as target analytes. The sorbent was characterized by cyclic voltammetry, scanning electron microscopy and Fourier transform infrared spectroscopy. After extraction and elution, the analytes were analyzed by a Thermo U3000 HPLC system. Several affecting parameters, including the volume of elution solution, sample pH value, sample flow rate and sample volume, were evaluated and optimized. Results The proposed method showed good linearity with low limits of detection (from 0.5 to 0.8 ng·mL−1) and ideal accuracy with spiked recoveries from 81.30% to 99.16%. A good enrichment factor (more than 200‐fold) together with good sensitivity was obtained with this method. Analysis of resveratrol and polydatin in Polygonum cuspidatum samples by this method is efficient. Conclusion The method developed in this work exhibits several significant merits, including easy operation and high extraction efficiency, indicating that electrochemically prepared polymer sorbent is useful for sample pretreatment and analysis of traditional Chinese medicine samples.
