Quantification of ferritin bound iron in human serum using species-specific isotope dilution mass spectrometry

Abstract: Ferritin is a hollow sphere protein composed of 24 subunits that can store up to 4500 iron atoms in its inner cavity. It is mainly found in the liver and spleen but also in serum at trace levels. Serum ferritin is considered as the best single indicator in assessing body iron stores except liver or bone marrow biopsy. However, it is confounded by other disease conditions. Ferritin bound iron (FBI) and ferritin saturation have been suggested as more robust biomarkers. The current techniques for FBI determinatio… Show more

“…There are different ways of mobilizing iron from cellular ferritin depending on the condition, and among others, a significant place in vivo is attributed to the decomposition of ferritin, the process of ferritinophagy (a subtype of autophagy). Serum ferritin contains less iron per molecule 27 , is largely composed of L-subunits in contrast to cellular ferritin. In contrast to the clinical potential of serum ferritin, the function or possible effects of serum ferritin are largely unknown at present.…”

“…Попав в цитоплазму, железо доставляется в различные внутрикле-точные участки, включая митохондрии для биосинтеза гема, и к ферритину -внутриклеточному депо железа [36]. Метаболически неактивное железо хранится в ферритине и находится в равновесии с обменным железом, связанным с молекулами-носителями [37].…”

“…True assessment of the utility of FBI requires development of more reliable, sensitive and practical technologies, which remove the difficulty of contamination and loss that may be introduced during a separation step. One recent publication attempts to mitigate this, using species-specific isotope dilution mass spectrometry (SS-IDMS) [110]. In SS-IDMS a sample of apoferritin is spiked with a known quantity of 57 Fe prior to purification.…”
