Quality Evaluation of Polar and Active Components in Crude and ProcessedFructus Corniby Quantitative Analysis of Multicomponents with Single Marker

Abstract: Objective. To develop a quantitative analysis of multicomponents by single-marker (QAMS) method for the simultaneous determination of polar active components in Fructus Corni. Methods. Loganin was selected as the internal reference, and the relative correction factors (RCFs) of gallic acid, 5-hydroxymethyl-2-furfural, morroniside, sweroside, cornin, 7α-O-methylmorroniside, 7β-O-methylmorroniside, 7α-O-ethylmorroniside, 7β-O-ethylmorroniside, and cornuside were established. The contents of multicomponents were … Show more

“…Secoiridoids are believed to be even more advanced due to the more complicated biostynthetic steps (Xiang et al, 1996). Thus, the most closely related in the iridoids transformation pathway compounds, such as loganin ( 1 ), loganic acid ( 3 ), cornin ( 4 ), sweroside ( 5 ), and representative of secoiridoids, cornuside ( 6 ), have been detected both in fruits of C. mas and C. officinalis (Deng et al, 2013; Ma et al, 2014; Perova et al, 2014; Sochor et al, 2014; Kucharska et al, 2015; Jiang et al, 2016). However, it should be noted that in the 26 cultivars of C. mas it was shown that the most abundant iridoid was loganic acid ( 3 ) (81.5–461.1 mg/100 g fw) constituting 88–96% of total iridoids (86.9–493.7 mg/100 g fw).…”

“…The relative correction factor for component I (internal standard) is the ratio of the absolute correction factor for component I to standard substance S (reference compound). In previous studies, loganin has also been used as the internal standard to calculate different components of Fructus corni (Jiang et al, ). Several studies on relative correction factors are available (Dou et al, ; He et al, ; He, Liu, Tian, Li, & Yang, ; Huang, Yang, Shi, Wang, & Wang, ; Kong, Zhu, Wang, Zhang, & Feng, ).…”

“…In previous studies of QAMS, researchers have investigated the stability of different columns and instruments (Wang, Zhang, Sun, & Chen, ). Other studies have investigated the reproducibility of the relative correction factors of different instruments and columns (Li et al, ; Zhu et al, ), and different column temperatures and flow rates (Jiang et al, ). In this study, the stability of the relative correction factors of different column temperatures (25, 30 and 30°C), two brands of columns (namely, Waters Acquity BEH C 18 , 100 × 2.1 mm, 1.7 μm and Thermo Syncronis C 18 , 100 × 2.1 mm, 1.7 μm) and three assistants were investigated.…”

“…Three components have clear chemical structure, obvious pharmaceutical properties, and convenient detection, so they should be considered for study first. QAMS, which has many advantages such as low cost and high efficiency, has been widely used in the determination of traditional Chinese medicine and multi-index components in recent years [ 17 , 22 – 26 ]. In this experiment, the 3 components to be tested were curcumin derivatives and curcumin was a representative component of these compounds and is easy to obtain.…”
