Quality evaluation for Radix Astragali based on fingerprint, indicative components selection and QAMS

Abstract: Radix Astragali (RA) is one of the most widely used Chinese herbs prescribed in many Chinese formulas to reinforce 'Qi' and treat vital energy deficiency. This study combined fingerprinting with quantitative analysis multi-components by a single marker (QAMS) to improve the quality control standard for RA on the basis of existing quality control methods of traditional Chinese medicinal materials. UPLC-ESI-TOF-MS technique was used to evaluate the quality of RA by fingerprinting and QAMS. Using the anti-inflamm… Show more

“…Methods for calculating the RCFs have been previously reported [24,37]. First, gastrodin was selected as the internal standard, and a multipoint method (Equation (1)) was used to calculate the relative correction factors (RCF) for p -hydroxy benzaldehyde, p -hydroxybenzyl alcohol, parishin A, parishin B, parishin E, and parishin C. Then the content of the measured component was calculated according to Equation (2) [38].…”

“…Moreover, it is stable, is easily obtainable, and can be well separated under the conventional chromatographic conditions. Using vitexin as a reference substance, f x was calculated by the ratio of the peak area and the concentration between the analyte and vitexin (see ( 1 )) [ 23 25 ]. Based on the specific f x of objective compound, its concentration ( C x ) in the sample solution could be calculated according to the following formula (see ( 2 )).…”

“…Comparing with ESM, QAMS reduces the expenditure on reference substances and shortens the operation time, especially suitable for quantification of a serial of analogues [ 22 ]. QAMS method has been used for the quality control of some herbal drugs, such as Panax notoginseng [ 23 ], Andrographis herba [ 24 ], and Radix astragali [ 25 ].…”

“…The acute inflammatory model of mice was based on previous reports with some modifications (Huang et al, ; Xiao et al, ). Mice were randomly divided into 12 groups with 10 mice per group: (a) the vehicle control group (mice treated with saline at an equal volume); (b) the model group (mice with inflammation treated with saline at an equal volume); (c) the positive control group [mice with inflammation treated with dexamethasone acetate tablets (3.12 mg/kg, V = 20 mL/kg)] and (d) the nine drug groups [mice with inflammation treated with GGD (S1–S9) at a clinically equivalent quadruple dose (1.48 mg/mL, V = 20 mL/kg) once a day for 3 consecutive days].…”
