Quality control tests of two salmonella enrichment media using different inocula

Abstract: SUMMARYInocula of polluted water naturally infected with salmonellas effectively distinguished six brands of selenite and six brands of tetrathionate enrichment media into satisfactory and unsatisfactory categories. Minimal inocula of pure cultures differentiated the tetrathionates, but not the selenites. Inocula of naturally infected chicken giblets suggested that there was a difference between two comparable brands of tetrathionate, but this was not statistically significant. The difference was, however, cle… Show more

“…It would not allow multiplication of small numbers of salmonellas (Harvey, Price & Crone, 1975). The Cardiff tetrathionate broth, incubated at 43°C would, however, permit growth of minimal numbers of salmonellas as would our seitz-filtered selenite F broth incubated at the same temperature.…”

“…Once prepared it is ready for use for at least one month. It is also cheap and, unlike some commercial enrichment media (Harvey, Price & Crone, 1975), the different laboratory-prepared lots are uniform in performance, provided malachite green oxalate (Merck) p.a. and MgCl2 .…”

“…Harvey and Price (1975) showed that the performance of different medium brands can vary depending on the inoculation concentration of Salmonella. Inoculation concentration also affects the lag-time of Salmonella Typhimurium (Stephens et al, 1997).…”
