Quality and Safety of Platelet Apheresis Concentrates Produced with a New Leukocyte Reduction System

Abstract: LRS apheresis appears to be a safe procedure, which produced effective PCs with few contaminating leukocytes. With new apheresis technology, filtration of PCs may become superfluous.

“…However, in both groups, the procedures resulted in comparably significant decreases in platelets, lymphocytes and monocytes. In similar apheresis procedures, data with a significant reduction of platelets and leucocytes have also been described for plateletpheresis [15,19]. These results agree, at least in part, with those of Nguyen et al who used the MNC program in melanoma patients [7].…”

“…wherein CE = collection efficiency, the yield represents the total number of cells in the leukapheresis product, PBV is the processed blood volume in µl (processed volume – citrate volume), precount is the donor's peripheral blood count (per µl) before leukapheresis, and postcount is the donor's peripheral blood count (per µl) after leukapheresis [15]. The processed part of the total blood volume (TBV in ml, as determined by the COBE Spectra™, based on the donor's gender, height and weight) was calculated according to the following formula:…”

“…After development of new leukoreduction systems 12,13 and incorporation of limits to WBC counts per therapeutic blood unit by the German and European Guidelines, 4,5 the effectiveness of leukoreduction is of high interest. The number of remaining WBCs within the TA products was significantly higher than with the AC platform, which confirms the observation of others 9 .…”

“…The system produces leukoreduced SDPs and plasma by a WBC reduction system (LRS). The operational principles of this system have recently been explained 2,3 . In the event of problems occurring during the procedure that could affect the separation interface, and therefore compromise the leukoreduction of the product, there is a software alarm that is similar to that previously described for the Trima system.…”
