Leukapheresis for the extraction of monocytes and various lymphocyte subpopulations from peripheral blood: product quality and prediction of the yield using different harvest procedures

Abstract: Leukapheresis is a safe and efficient procedure for collecting large numbers of peripheral blood monocytes and different lymphocyte populations from non-mobilized donors. The two programs yield comparable leucocyte harvests. Based on our results, yields can be predicted from the peripheral cell counts.

“…The role of monocyte transfusions in these patients has not been explored despite leukapheresis having been recently shown to be a safe and efficient procedure for collecting large numbers of peripheral blood monocytes from donors [23]. Awareness of the complimentary effect between platelets and monocytes in reducing postoperative blood loss may thus be of use in developing new strategies for the management of patients having postoperative bleeding diatheses not controlled by standard measures.…”

“…This is not only possible for healthy donors but can be achieved with patients as well [40][41][42]. The collected mononuclear cells are harvested under the same conditions as classical blood products such as thrombocytapheresis products and are suitable for further processing in GMP facilities.…”

“…2) were sealed as far as possible from the side of the channel. The ''blood in'' tube (red) (1) was sterile connected to an empty 1 L transfer bag using the Terumo Sterile Tubing Welder (SC-201H, Terumo Company, Japan) whereas the ''plasma out'' tube (transparent) (6) was sterile connected to a saline containing transfer bag. By gravity, the cells were washed with the saline and transferred to the empty bag.…”
