Milk is a natural fluid and as such contains small amounts of naturally occurring steroids. Human milk is recommended as the optimal source of nutrients for infants and young children, and it has been associated to several short- and long-term benefits. For this reason, its composition is used as a reference for designing infant formulas. However, the available information on the hormonal levels of these dairy products is scarce, and it is usually limited to estradiol and estrone. In the present study, six natural sex hormones (pregnenolone, progesterone, estrone, dehydroepiandrosterone, testosterone and androstenedione) have been extracted from sixteen milk-based infant formulas and analyzed with liquid chromatography tandem mass spectrometry (HPLC-MS/MS). The purpose of this research was to quantify natural steroid hormones in various infant formulas, to provide food and nutrition practitioners with information to estimate intakes in children. In addition, data found in the literature was used for comparison. The findings suggest that there are certain similarities between bovine milk and dairy products for infants. Furthermore, the detected levels were in general lower than those observed in human milk and/or colostrum. The reported results represent a valuable addition to the current knowledge on natural hormone content of infant foods.

a b s t r a c tA novel method for simultaneous determination of five estrogens and four androgens by online solid-phase extraction (SPE) coupled with high-performance liquid chromatography-tandem mass spectrometry (HPLC-ESI-MS/MS) in water samples was developed. An aliquot of 50 mL water sample after filtration was injected directly into autosampler and the analytes were preconcentrated on a NG1 online SPE column. After cleanup step the analytes were eluted in back flush mode and then separated on a liquid chromatography column. The experimental parameters, such as sample loading flow rate, cleanup condition and elution time, were optimized in detail. Estrogens and androgens were detected in negative and positive mode, respectively. High ionization efficiency of all the analytes was achieved by adding of 1‰ ammonia in the mobile phase. The recoveries ranged from 31.8% to 119.0% and the inter-day RSDs ranged from 2.7% to 19.6%. The limits of detections (LODs) were between 0.1 and 2.5 ng/L. The proposed method was successfully applied to the analysis of three types of water samples, including river water, influent and effluent water from a wastewater treatment plant (WWTP). The recoveries of androgens were not that good and a further study is being planned to improve the sensitivity for them. The proposed method is simple, sensitive and suitable for simultaneous analysis and monitoring of estrogens and androgens in water samples.

Bovine somatostatin is detectable in milk; however, there is no evidence that the protein persists in the environment nor that it is active in humans. In contrast, steroids are transported through milk and meat to humans where they may exert biological activity. Furthermore, environmental matrices such as raw water and dust may also allow for the environmental transport and bioavailability of steroids to humans. Endogenous and exogenous steroids can be found in the meat, milk, and waste materials produced by cattle. While the concentrations may be low, exposure to these matrices, most notably dairy products made with whole milk, can be a source of exogenous steroids to humans.
