Late blight (Phytophthora infestans) can have devastating effects on tomato production over the whole world. Most of the commercial cultivars of tomato, Solanum lycopersicum, are susceptible. Qualitative and quantitative resistance has been described in wild relatives of tomato. In general qualitative resistance can more easily be overcome by newly evolved isolates. Screening of three S. habrochaites accessions (LA1033, LA2099 and LA1777) through a whole plant assay showed that accession LA1777 had a good level of resistance to several isolates of P. infestans. To explore the potential in this wild species, an introgression line (IL) population of S. habrochaites LA1777 was used to screen individual chromosome regions of the wild species by a detached leaf assay. Two major isolates (T 1,2 and T 1,2,4 ) were used and two parameters were measured: lesion size (LS), and disease incidence (DI). Substantial variation was observed between the individual lines. QTLs were identified for LS but not for DI.The presence of five QTLs derived from LA1777 (Rlbq4a, Rlbq4b, Rlbq7, Rlbq8 and Rlbq12) results in unambiguous higher levels of resistance. All QTLs co-localized with previously described QTLs from S. habrochaites LA2099 except QTL Rlbq4b, which is therefore a novel QTL.

Enzymes interact to generate metabolic networks. The activities of more than 22 enzymes from central metabolism were profiled during the development of fruit of the modern tomato cultivar Solanum lycopersicum 'M82' and its wild relative Solanum pennellii (LA0716). In S. pennellii, the mature fruit remains green and contains lower sugar and higher organic acid levels. These genotypes are the parents of a widely used near introgression line population. Enzymes were also profiled in a second cultivar, S. lycopersicum 'Moneymaker', for which data sets for the developmental changes of metabolites and transcripts are available. Whereas most enzyme activities declined during fruit development in the modern S. lycopersicum cultivars, they remained high or even increased in S. pennellii, especially enzymes required for organic acid synthesis. The enzyme profiles were sufficiently characteristic to allow stages of development and cultivars and the wild species to be distinguished by principal component analysis and clustering. Many enzymes showed coordinated changes during fruit development of a given genotype. Comparison of the correlation matrices revealed a large overlap between the two modern cultivars and considerable overlap with S. pennellii, indicating that despite the very different development responses, some basic modules are retained. Comparison of enzyme activity, metabolite profiles, and transcript profiles in S. lycopersicum 'Moneymaker' revealed remarkably little connectivity between the developmental changes of transcripts and enzymes and even less between enzymes and metabolites. We discuss the concept that the metabolite profile is an emergent property that is generated by complex network interactions.

ABSTRAKPenyimpangan segregasi marka biasa terjadi pada persilangan antarspesies pada berbagai komoditas tanaman. Penelitian terdahulu mengenai pemetaan lokus ketahanan terhadap jamur hawar dini (Alternaria solani) menunjukkan penyimpangan segregasi dari 52% marka yang terpetakan pada peta pautan 176 progeni F 2 dari persilangan Solanum lycopersicum cv. Solentos × S. arcanum LA2157. Tujuan penelitian ini adalah menganalisis lebih detil segregasi marka pada peta pautan tersebut dan menentukan penyebab penyimpangan segregasi dengan menghitung frekuensi alel dan genotipe F 2 dari tiap marka. Dari 371 marka yang terpetakan, 192 di antaranya menyimpang dari rasio Mendel 1 : 2 : 1. Penyimpangan marka terjadi pada semua kromosom, sebesar 1% sampai 9%. Surplus alel homozigot S. arcanum (40%) menjadi penyumbang terbesar penyimpangan segregasi, diikuti oleh heterozigot (18%) dan homozigot S. lycopersicum (5%). Frekuensi alel dari 152 marka menyimpang dari frekuensi homogenitas alel yang diharapkan, yang mungkin disebabkan oleh seleksi gametofitik. Enam puluh satu marka menyimpang dari distribusi frekuensi genotipe F 2 yang diharapkan, yang dapat diakibatkan oleh segregasi zigotik. Segregasi 37 marka menyimpang dari frekuensi homogenitas alel dan distribusi genotipe F 2 yang diharapkan. ABSTRACTDistorted marker segregation is a common phenomenon in interspecific cross of various crops. Previous mapping study of early blight fungus (Alternaria solani) resistance loci showed 52% marker distortion in the genetic linkage map of 176 F 2 progenies derived from Solanum lycopersicum cv. Solentos × S. arcanum LA2157. The objectives of this study were to analyze in detail the marker segregation in the map and to determine the cause of segregation distortion by calculating the allele and genotype frequencies of each marker. Out of 371 mapped markers, 192 markers deviated from the expected Mendelian ratio of 1 : 2 : 1. Distorted markers occurred in all chromosomes, ranging from 1% to 92%. Surplus of S. arcanum homozygotes contributed most to the skewness (40%), followed by heterozygotes (18%), and S. lycopersicum homozygotes (5%). The allele frequencies of 152 markers deviated from the expected allele homogeneity frequency, indicating that their segregation might be affected by gamethophytic selection. Sixty-one markers deviated from the expected F 2 genotype frequency distribution, indicating that their segregation might be influenced by zygotic selection.
