Purification Procedure for the Determination of Niacin Vitameres in Gastrointestinal Contents and Blood

Abstract

Summary A solid phase extraction procedure for subsequent simultaneous HPLC analysis of free nicotinic acid (NIA) and nicotinamide (NAM) in dried or liquid rumen and gastrointestinal contents from sheep is described. Twenty‐five mg dried samples were suspended in 1.0 ml of 0.1 mol/l Li2SO4 and centrifuged. The supernatants were passed through two 500 mg solid phase extraction columns (Bond Elut NH2 and Sep Pak C18), mounted in series, and eluted by a four step pH gradient procedure. Recoveries of the two vitameres were between 70 and 75 %. Purification was satisfactory when the method was applied to different fractions of rumen content (e. g. to food residues, to a microbial preparation and to rumen fluid), to intestinal contents of sheep and to blood. In rumen fluid no free NIA and NAM was found. Food particles and rumen microbes contained no free NAM. Zusammenfassung Eine Reinigungsmethode für die Bestimmung von Niacinvitameren aus Magen‐Darminhalt und Blut Für eine simultane Bestimmung von Nicotinsäure (NIA) und Nicotinamid (NAM) mit HPLC müssen viele biologische Proben zuvor intensiv gereinigt werden. Es wird eine Reinigungsmethode mit Festphasenextraktion für Pansen‐ und Darminhalt sowie für Blut von Schafen beschrieben. Getrocknete Proben (25 mg) wurden in 1,0 ml einer 0,l mol/l Li2SO4 Lösung suspendiert und zentrifugiert. Der überstand wurde auf zwei verbundene Festphasenextraktionssäulen (Bond Elut NH2 und Sep Pak C 18, je 500 mg Sorbensmittel) gegeben und mit einem stufenweisen pH‐Gradienten (pH 2,0; 2,5; 3,0 und 3,5) eluiert. Drei Fraktionen enthielten beide Niacinvitamere mit einer Wiederfindungsrate von etwa 70–75%. Die Reinigung war ausreichend für eine anschließende HPLC‐Bestimmung von NIA und NAM aus drei verschiedenen Fraktionen von Panseninhalt, aus Darminhalt oder aus Blut. In der partikelfreien Flüssigkeit des Pansens von Schafen, die mit einem Heu und Konzentrat gefüttert worden waren, war weder NIA noch NAM nachweisbar. Futterpartikel und Mikroorganismenfraktion enthielten NIA.
