a b s t r a c tThe hydrocolloid extracted from Caesalpinia ferrea var. ferrea seeds purified by membrane filtration was denominated GMPF (14% yield) and contained 75% total carbohydrate and 9% protein, and the mannose to galactose (Man:Gal) ratio was around to 2.1:1.0. The periodate oxidation, methylation and 1D/2D NMR spectra results indicated galactomannan (GM) with a (1 → 4)-linked ␤-d-mannopyranose backbone, partially substituted at O-6 with single-unit ␣-d-galactopyranose side-chains. The splitting of three 13 C signals in the regions of the 4-O-Manp units revealed that the ␣-d-Galp units were irregularly (54%) distributed at the main chain. By rheology, GPC-coupled to multi-detectors, static and dynamic light scattering it was observed that the GM in nitrite solution presented an intrinsic viscosity of 860 mL/g, a polydispersity of 1.55, and a structural parameter (R g /R h ) of 1.77, showing a random coil and flexible conformation, which was confirmed by the Mark-Houwink constant˛(0.63). The results suggest that GMPF can be a new source of GM.

Abstract:In this study the viability of using guar gum to form films was investigated along with the effectiveness of the cross-linking process employing 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) as the cross-linking agent. In addition, the cross-linked films were evaluated considering the water absorption, thermal stability and mechanical properties. The cross-linking process of guar gum films was confirmed by the low solubility in water and through infrared analysis. The results shown that the properties evaluated were affected by the cross-linking process due to changes in the polysaccharide structure. For example, the swelling behavior and water vapor absorption decreased with an increase in the amount of EDC. The EDC content (10-30%) also affected the polymer structure and hydrogen bond formation, reducing the thermal stability of the system.

Galactomannan in industrial Gleditsia microphylla and guar gum was successfully fractionated by gradual precipitation in an aqueous solution with increasing ethanol concentrations. The molecular properties of each fraction were characterized, and the galactomannans were added to photopolymerized hydrogels to test their effects on mechanical properties and swelling capacity. In the series fractions of guar gum, the sample precipitated from 20% EtOH solution had the highest yield, mannose to galactose ratio, and viscosity, and it had a slightly lower molecular weight than that precipitated by 30% EtOH. Correspondingly, the best tensile property of its photopolymerized hydrogel was finally detected. In terms of G. microphylla gum, the precipitation in 30% EtOH solution achieved the highest yield, M/G ratio, and molecular weight value, and it exhibited the best rheological property of all the samples. The hydrogel with the addition of this sample also had the best mechanical properties despite its lower hydroscopicity than the blank hydrogel. The unique properties of each fraction could probably lead to their use as biodegradable alternatives in different applications.
