Purification and sequence characterization of chondroitin sulfate and dermatan sulfate from fishes

Abstract: Chondroitin sulfate (CS) and dermatan sulfate (DS) were extracted and purified from skins or bones of salmon (Salmo salar), snakehead (Channa argus), monkfish (Lophius litulon) and skipjack tuna (Katsuwonus pelamis). Size, structural sequences and sulfate groups of oligosaccharides in the purified CS and DS could be characterized and identified using high performance liquid chromatography (HPLC) combined with Orbitrap mass spectrometry. CS and DS chain structure varies depending on origin, but motif structure … Show more

“…Skipjack tuna (Katsuwonus pelamis) is the main resource of canned fish and shares of more than 70% of tuna catches (3,26,27). Many researches have utilized by-products of skipjack tuna to produce some functional components, such as collagens and gelatins (28,29), BPs (9,21,30), oil and lipid (11,31), and multiple sulfated polysaccharides (32). Roe is one of the main by-products of skipjack tuna and is rich in crude proteins with the content of 21.00 ± 0.06% of tuna roe (on a wet weight basis) (33).…”

“…Where do these doping heteroatoms come from? Usually, tuna bones are mainly composed of collagen, chondroitin sulfate (CS), , protein, and HAP. Among them are CS with abundant sulfonic acid groups and amine groups, and HAP with phosphate groups, which leads to heteroatom doping of TBCs.…”

“…The Lophius litulon (monkfish) are mainly distributed in the north-east Atlantic and live in a wide bathymetric range from shoal to seafloor of at least 1,000 m (Laurenson & Priede, 2005). As an abyssal fish with high nutrition and active substance content, the Lophius litulon have been wildly used for food and nutrition, including as a source bioactive substances and biological material (Lin et al, 2017;Stollewerk et al, 2014;Swart et al, 2019;Vázquez et al, 2020). The fish by-products from Lophius litulon processing could reach about 60% of the total weight with the liver occupying about 10%.…”
