Purification and Characterization of Antioxidant Peptides Derived from Protein Hydrolysate of the Marine Bivalve Mollusk Tergillarca granosa

Abstract: In this report, protein hydrolysate (TGH) of blood cockle (Tegillarca granosa) was prepared using a two-enzyme system (Alcalase treatment for 1.5 h following Neutrase treatment for 1.5 h). Subsequently, six antioxidant peptides were isolated from TGH using ultrafiltration and chromatography methods, and their amino acid sequences were identified as EPLSD, WLDPDG, MDLFTE, WPPD, EPVV, and CYIE with molecular weights of 559.55, 701.69, 754.81, 513.50, 442.48, and 526.57 Da, respectively. In which, MDLFTE and WPPD… Show more

“…The structure-activity relationship of the functional molecule will provide the basis for predicting the activity of new compounds, but so far it is still difficult to illuminate the relationship between structural characteristics and antioxidant activity of peptides [5,46]. In general, molecular size, hydrophobicity, and amino acid composition and sequence are regarded as key factors for the activity of antioxidant peptides [47,48].…”

“…Two peptide hydrolysate fractions from the Marine Bivalve Mollusc, Tergillarca granosa, have also been demonstrated to exhibit antioxidant activity [57]. The peptides with amino acid composition (and thus designated) MDLFTE and WPPD exhibited strong DPPH free radical scavenging (EC50 = 0.53 and 0.36 mg/mL, respectively), hydroxyl radical scavenging (EC50 = 0.47 and 0.38 mg/mL respectively), superoxide anion radical (EC50 = 0.75 and 0.46 mg/mL, respectively) and ABTS cation radical scavenging (EC50 = 0.96 and 0.54 mg/mL, respectively) activities, although these antioxidant activities were not sustained at basic pH conditions (pH > 9 for 2.5 h), high temperatures (>80ºC for 0.5 h) nor during simulated GI digestion [57].…”

“…In recent years, seafood-derived peptides with antihypertensive, antioxidant, and antitumor activities have been characterized from many marine products and their by-products [10,11,[20][21][22]. In contrast, little attention has been paid to peptides with hypolepidemic, anti-obesity, antiatherogenic and cardioprotective effects [2].…”
