Helicobacter pullorum is an emerging foodborne pathogen that commonly colonizes the gastrointestinal tract of poultry, causing gastroenteritis. It has been related to several clinically important infections, including colitis and hepatitis, inflammatory bowel disease, recurrent diarrhea, and bacteremia in the human population. The bacterium may be transmitted to humans through undercooked poultry meat. In order to investigate the occurrence of H. pullorum in raw retail chicken meat (thighs and breasts), we analyzed 240 samples: 120 chicken thigh and 120 chicken breast samples. The samples were analyzed by means of an isolation protocol using Steele and McDermott’s modified filtration technique on Brucella agar supplemented with 5% of defibrinated sheep’s blood. The presumptive colonies were biochemically identified and analyzed using a previously described conventional PCR test based on the 16S rRNA gene. In total, 35% of analyzed samples were positive using the microbiological protocol and 45% were positive by PCR. These results suggest that H. pullorum can be transmitted to humans through the handling and consumption of raw poultry meat, representing a risk for food business operators and consumers. Efforts to control H. pullorum in broiler meat should prioritize the implementation of stringent hygienic practices across all stages of the food chain, from the farm to the consumer.

Decreased vessel wall function and elasticity lead to intracranial aneurysms (IAs), which increase the risk of vessel rupture. The association between IA formation and gut flora remains unclear. Therefore, we compared the gut microbiomes of patients with IAs with those of healthy controls (HCs) to identify specific bacteria associated with IAs. This single-center prospective case-control study was conducted from July 2022 and February 2023. Stool samples were collected from patients with unruptured IAs and HCs. The bacterial communities of patients with IAs were compared with those of healthy controls using 16SrRNA sequencing technology. In addition, phylogenetic trees were constructed to identify specific species. Twenty-five patients with IAs and 30 HCs were included in this study. There were no significant differences in age, sex, body mass index, hypertension, diabetes mellitus, dyslipidemia, smoking, or alcohol consumption rates between the two groups. Diversity analysis indicated no significant difference in alpha diversity; however, there was a significant difference in beta diversity with respect to the unweighted UniFrac distance. Linear discriminant analysis combined with effect size measures showed that several intestinal microbiome taxonomic profiles were significantly enriched in samples from patients in the IA group. Helicobacter species were our point of concern as they are widely considered pathogenic. Thus, we constructed a Helicobacter-related phylogenetic tree and identified three species, Helicobacter pullorum, Helicobacter typhlonius, and Helicobacter rodentium. The gut microbiome profiles of patients with stable, unruptured IAs differed significantly from those of HCs. Therefore, the genus Helicobacter may be associated with IA formation.

Even though Helicobacter pylori (H. pylori) is a serious pathogen, its origin is unknown. Poultry (chicken, turkey, quail, goose, and ostrich) is consumed as a regular protein source by many people across the world; therefore, sanitary ways of delivering poultry for food are important for global health. As a result, the distribution of the virulence genes cagA, vacA, babA2, oipA, and iceA in H. pylori isolates in poultry meat, as well as their antibacterial resistance, was investigated. A Wilkins Chalgren anaerobic bacterial medium was used to cultivate 320 samples of raw poultry meat. Disk diffusion and multiplex-PCR were used to investigate both antimicrobial resistance and genotyping patterns. H. pylori was found in 20 of 320 (6.25 %) raw chicken meat samples. The highest incidence of H. pylori was found in chicken raw meat (15%), whereas no isolate was recovered from goose or quail raw meat (0.00%). Resistance to ampicillin (85%), tetracycline (85%), and amoxicillin (75%) were the most commonly detected in the tested H. pylori isolates. The percentage of H. pylori isolates with a multiple antibiotic resistance (MAR) index value of more than 0.2 was 17/20 (85%). The most prevalent genotypes detected were VacA (75%), m1a (75%), s2 (70%) and m2 (65%), and cagA (60%). The most typically detected genotype patterns were s1am1a (45 %), s2m1a (45 %), and s2 m2 (30 %). babA2, oipA+, and oipA- genotypes were found in 40%, 30%, and 30% of the population, respectively. In summary, fresh poultry meat was polluted by H. pylori, with the babA2, vacA, and cagA genotypes being more prevalent. The simultaneous occurrence of vacA, cagA, iceA, oipA, and babA2 genotypes in antibiotic-resistant H. pylori bacteria raises a serious public health concern regarding the consumption of raw poultry. Future research should evaluate antimicrobial resistance among H. pylori isolates in Iran.
