PTX3, A Prototypical Long Pentraxin, Is an Early Indicator of Acute Myocardial Infarction in Humans

Abstract: PTX3 is present in the intact myocardium, increases in the blood of patients with AMI, and disappears from damaged myocytes. We suggest that PTX3 is an early indicator of myocyte irreversible injury in ischemic cardiomyopathy.

“…The blood was processed to obtain plasma and serum samples that were immediately frozen and stored at Ϫ70°C until analysis. The determinations were made in a single run by central laboratory personnel blinded to the experimental groups and included brain natriuretic peptide (BNP, MEIA AxSYM-Abbott, Abbott Park, IL), high-sensitivity cardiac troponin-T (hs-cTnT, Elecsys, Roche Diagnostics, Mannheim, Germany) 16 , pentraxin-3 (PTX3, ELISA based on in-house reagents 17 ), high-sensitive C-reactive protein (hsCRP, turbidimetric method, Beckman Coulter DxC800, Brea, CA), myoglobin (MEIA AxSYM-Abbott), creatine kinase (CK, Rosalki enzymatic method), creatine kinase isoenzyme MB (CK-MB, MEIA Axsym-Abbott, Abbott Park, IL). Urinary albumin excretion was measured using a radioimmunoassay (coated tubes, Immunotech, Marseille, France), indexed to creatinine, and expressed as the urinary albumin-to-creatinine ratio (UACR, mg/g of creatinine).…”

“…The levels of PTX3 in plasma or culture supernatant were measured by ELISA, based on mAb MNB4 and rabbit antiserum, as previously described (18). For cell culture experiments cells were grown to confluence in 24-well plates, serum-starved overnight, and then stimulated using DMEM containing 0.25% BSA.…”

“…This is supported by studies showing the ability of PTX3 to bind to the C1q component of the complement cascade (12) and to participate in the clearance of apoptotic cells (16). Moreover, PTX3 is elevated in critically ill patients, with a gradient from systemic inflammatory response syndrome to septic shock (17), and in several other diseases, such as myocardial infarction (18), rheumatoid arthritis (19), atherosclerosis (20), and small vessel vasculitis (21).…”

“…It turned out that there was an association of plasma pentraxin 3 concentration and carotid atherosclerosis found in the control group of patients with cardiovascular complications but with normal glucose tolerance. In the group of patients with type 2 diabetes and cardiovascular complications we have found an association of plasma concentration of PTX3 is not only the early marker of irreversible cardiac damage, but can also predict its future course [8,9]. Moreover, Hudzik et al proved that PTX3 may be a potential early marker of cardiovascular risk before the increase of systemic markers like hsCRP [10].…”
