Protoplasts: a friendly tool to study aluminum toxicity and coffee cell viability

Abstract: ObjectiveAluminum toxicity is a major limiting factor with regard to crop production and quality in most acidic soils around the world. We propose the use of C. arabica L. protoplasts to evaluate the toxic effects of aluminum, the nuclear localization of aluminum and propensity of aluminum to cause DNA damage.ResultsAfter protoplasts were exposed to aluminum (Al) for varying periods of time (0, 5, 10, 20 and 30 min), we detected a reduction in protoplast viability. Additionally, we observed a rapid decline in … Show more

“…The MDA content in date palm callus treated with Al at 200 mg.l -1 was increased by up to 1.71-fold compared with that in the control callus. Lipid peroxidation was also induced under Al stress [14, 19, 27, 35]. The increase in lipid peroxidation may be attributed to the binding of Al to biomembrane and leads to rigidity, which in turns causes the generation of radical chain reactions by Fe ions [21, 48].…”

“…Al 3+ ions are genotoxic and cause direct damage to DNA [ 69 ]. Sumoylation involves small ubiquitin-like modifiers (SUMOs), which are attached to or detached from proteins to modify their function and subcellular localization.…”

“…DNA damage evaluation is another frequently employed marker to assess protoplast viability or the effect of genotoxicity of environmental pollutants and abiotic stresses on protoplasts. In these procedures, protoplasts are used as a direct source of nuclei to perform gel electrophoresis with ethidium bromide staining in order to detect DNA laddering ( Poot-Poot et al, 2016 ). Identically, single cell gel electrophoresis assay (SCGE), also called Comet assay, allows the study of DNA damage on protoplasts at the single cell or nuclei level ( Kuzminsky et al, 2016 ; Badaró Costa et al, 2018 ; Choury et al, 2018 ).…”
