Protocol for ethyl methanesulphonate (EMS) mutagenesis application in rice

Abstract: Background: Non-transgenic chemical mutagen application, particularly ethyl methanesulfonate (EMS), is an important tool to create mutations and gain a new genetic makeup for plants. It is useful to obtain a sufficient number of mutant plants instead of working with a severe mutation in a few plants. EMS dose and exposure period have been previously studied in several crops; however, EMS used to create point mutations in presoaked rice seeds has not been sufficiently studied and there is no standard protocol f… Show more

“…Typically, 500 seeds m -2 are sown. The seeds were soaked in water for 48 hours for pre-germination (Unan et al, 2022), and seeding was performed in wet soil conditions. Pre-germinated seeds were directly sown by hand in the flooded field.…”

“…Thai highland rice (cv. Dawk Pa-yawm and Dawk Kha 50) was subjected to induced mutagenesis using EMS to create genetic variability ( Awais, Nualsri & Soonsuwon, 2019 ; Unan et al, 2022 ). By leaving out this stage, the mutagen dose can result in either a high or low mutation frequency ( Barr & Fearns, 2016 ; Espina et al, 2018 ; Galal & Thabet, 2018 ).…”
