Proteomic profiling of retinoblastoma by high resolution mass spectrometry

Abstract: BackgroundRetinoblastoma is an ocular neoplastic cancer caused primarily due to the mutation/deletion of RB1 gene. Due to the rarity of the disease very limited information is available on molecular changes in primary retinoblastoma. High throughput analysis of retinoblastoma transcriptome is available however the proteomic landscape of retinoblastoma remains unexplored. In the present study we used high resolution mass spectrometry-based quantitative proteomics to identify proteins associated with pathogenesi… Show more

“…IGF2BP1 was showed to be overexpressed in both retinoblastoma and choriocarcinoma. In retinoblastoma, the gene promotes tumor cell proliferation and migration [ 99 ]. In choriocarcinoma cell lines, the depletion of ribosomal protein S6 kinase (RSK2) or protein phosphatase methylesterase 1 (PPME1) inhibits cell migration and invasion, which is similar to that after knockdown of IGF2BP1 that controls RSK2 and PPME expression, whereas it did not influence cellular proliferation and morphology [ 100 ], indicating that IGF2BP1 promotes choriocarcinoma cell migration and invasion partly via the effect of RSK2 and PPME1.…”

“…Besides, proteins and non-coding RNAs, specifically miRNAs of exosomes, are identified, isolated, and characterized for their role in various pathological conditions [ 93 , 94 ]. In the recent study, the proteins of exosomes are characterized by the RB tumor tissue and from the RB vitreous seeding (RBVS) in the vitreous humor [ 95 ]. In RBVS exosomes, the proteins involved in the invasion and metastasis are identified, regulating functions, such as extracellular matrix (ECM) remodeling and metabolism of glucose and amino acids, etc.…”

“…In RBVS exosomes, the proteins involved in the invasion and metastasis are identified, regulating functions, such as extracellular matrix (ECM) remodeling and metabolism of glucose and amino acids, etc. [ 95 ]. Besides, to prognosticate RB, less invasive procedures, such as a liquid biopsy, would serve as the best option for children and are less painful than the invasive procedures, such as bone marrow aspiration or lumbar puncture, for the detection of metastasis.…”

“…In this section, we detail how these datasets could be utilized to make testable predictions into the molecular reprograming of Retinoblastoma cells. Microarray transcriptional profiling and proteomics analysis of Retinoblastoma tumors have identified a number of putative metabolic changes within the tumor cells ( Kooi et al, 2015 ; Danda et al, 2016 ). From these studies, dysregulated lipid metabolism, mitochondrial energy metabolism and photoreceptor metabolism were implicated as key processes linked to Retinoblastoma progression.…”
