Proteomic analysis of somatic embryogenesis in Vitis vinifera

Abstract: Two dimensional gel electrophoresis coupled to mass spectrometry has been used to study the somatic embryogenesis in Vitis vinifera, by comparing embryogenic and non embryogenic calluses of the Thompson seedless cv. More than 1,000 spots were reproducibly resolved in colloidal Coomassie brilliant blue stained gels over a pI nonlinear range of 3-10 in the first dimension and using homogeneous 12.5% polyacrylamide gels in the second dimension. The expression pattern of 35 spots differed significantly between the… Show more

“…The characterization of profiles and changes in protein content during seed development allows for comparing in vitro and in vivo embryo development. However, although several embryogenically expressed proteins have been identified so far, there have been few reports focused on woody plants (Marsoni et al 2008), even less so regarding proteomic characterization throughout recalcitrant seed development (Pawlowski et al 2007;Silveira et al 2008;Balbuena et al 2009). …”

“…Total proteins (500 mg) were loaded onto an 18-cm, pH 4-7 linear gradient or pH 3-10 nonlinear gradient (NL) Immobiline DryStrip gels (IPG strips) (GE Healthcare, Uppsala, Sweden), and separated as described by Marsoni et al (2008). The SDS-PAGE gels were visualised by the modified Colloidal Coomassie Brilliant Blue staining method (Aina et al 2007).…”

“…persicum (Lyngved et al, 2008;Winkelmann et al, 2006), V. vinifera (Marsoni et al, 2008) e Citrus sinensis (Pan et al, 2009). Em A. angustifolia, esta abordagem foi estabelecida e empregada, sendo identificadas uma série de proteínas marcadoras de estádio específicos durante os processos do desenvolvimento do embrião zigótico e germinação da semente (Balbuena et al, 2009dos Santos et al, 2006;Silveira et al, 2008;).…”
