Protein corona composition of poly(ethylene glycol)- and poly(phosphoester)-coated nanoparticles correlates strongly with the amino acid composition of the protein surface

Abstract: Extensive molecular dynamics simulations reveal that the interactions between proteins and poly(ethylene glycol) (PEG) can be described in terms of the surface composition of the proteins. PEG molecules accumulate around non-polar residues while avoiding the polar ones. A solvent-accessible-surface-area model of protein adsorption accurately fits a large set of data on the composition of the protein corona of poly(ethylene glycol)- and poly(phosphoester)-coated nanoparticles recently obtained by label-free pro… Show more

“…We are just starting to predict the proteins adsorbed and consequently the biological impacts. [142] Here,n anocarriers with one highly predominant protein help to gain af irst understanding.T his has recently been demonstrated for explaining the "stealth effect". Nanocarriers as well as technical surfaces are readily recognized by the immune system as proteins such as immunoglobulins or complement factors adsorb to these surfaces.T hen immune cells try to eliminate these marked foreign substances.M odifications of surfaces have been established that do not result in immune recognition by these mechanisms-this has been termed the "stealth effect/property".…”

“…Es wurden Modifikationen an Oberflächen entwickelt, die nicht zu einer Immunerkennung durch diese Mechanismen führen -d ies wurde als "Stealth-Effekt/Eigenschaft" bezeichnet. [142] Hiermit kann die Liste der adsorbierten Proteine korrekt vorhergesagt werden. Proteinabweisende und Anti-Zellaufnahme-Eigenschaften wurden PEG als Beschichtungsmaterial zugeschrieben.…”
