Engineering Proteins at Interfaces: From Complementary Characterization to Material Surfaces with Designed Functions

Abstract: Once materials come into contact with a biological fluid containing proteins, proteins are generally-whether desired or not-attracted by the material's surface and adsorb onto it. The aim of this Review is to give an overview of the most commonly used characterization methods employed to gain a better understanding of the adsorption processes on either planar or curved surfaces. We continue to illustrate the benefit of combining different methods to different surface geometries of the material. The thus obtain… Show more

“…4 Combining the thermodynamic information provided by ITC experiments with information from other methods gives deep insight into the mechanism of NP-protein interactions and the fate of protein-covered nanomaterials. 5 It is not the aim of this short review to reflect the addressed topics in their entirety, but to focus on the fundamental aspects of ITC, which can be important to the field of protein corona research. Furthermore, limitations of the method's state of the art and the use of auxiliary techniques for overcoming those limitations are addressed.…”

“…[13] However, the question is whether truly significant amounts of protein remains on the surface of every nanocarrier.L ooking at all these aspects, adeeper understanding of the biological impact and importance of the hard as well as the soft protein corona is necessary to proceed in the field of nanomedicine.T herefore,i nt his Minireview,w eg ive an overview about how the sample preparation by commonly established separation methods-that is the separation of nanocarrier-protein complexes from the surrounding medium-influences the outcome of the protein corona analysis.Special focus was placed on the requirements and limitations of the sample preparation process itself as the different characterization techniques used subsequently have already been reviewed extensively. [8,12,[14][15][16] One of the biggest challenges in the field of nanomedicine is the adsorption of biomolecules on the nanomaterial upon contact with abiological medium. The interactions of the resulting protein corona are essential for their behavior in abiological system.…”
