Protein analysis with terbium (III) and sodium dodecyl sulphonate by a second‐order scattering technique

Abstract: This is the first report of terbium(III) as a probe of second-order scattering (SOS) for the determination of proteins in human serum at nanogram levels. A sensitive method has been developed using light scattering, based on the fact that the weak SOS of proteins can be enhanced in the presence of terbium(III) and sodium dodecyl sulphonate (SDS). With this method, 7.0 x 10(-9)-1.0 x 10(-5) g/mL human serum albumin (HSA) and 5.0 x 10(-9)-5.0 x 10(-6) g/mL gamma-globulin can be determined; the detection limits w… Show more

“…Similarly, emission occurs at 270 nm and 267 nm (instead of 275 nm and 269 nm) when excitation was set up at 210 nm and 207 nm respectively. By reviewing the previous values of λ em upon conducting SFS, it could be concluded that SFS result at emission wavelength=λ ex +Δλ [12,13]; i.e. 225+60= 285 nm, 214+60=274 nm, 210+60=270 nm, and 207+60=267 nm.…”

“…Constant wavelength synchronous fluorescence spectroscopy (CWSFS) is an advanced fluorescence method capable of performing multi-component analysis, where simple spectra characterized by sharp narrow bands result [12,13]. This capability is attributed to Rayleigh scattering elimination which assists in getting over the spectral overlap experienced in normal spectrofluorometry [12,13].…”

“…The qualitative and quantitative analysis of protein is important in biochemistry, clinical medical science and food examination. Up to now, there have been several methods for the determination of total protein, such as spectrophotometry (1)(2)(3), resonance light-scattering (4)(5)(6)(7)(8), fluorescence (9)(10)(11)(12), electrochemistry (13) and luminescence methods (14). Among these, the fluorimetric method has been widely used because it has high sensitivity, convenient performance and good selectivity.…”

“…Recently, metal complexes as RLS probes have been used for the determination of protein (22,23). Rare-earth elements play an important role in agriculture and animal husbandry, clinical diagnosis and treatment, etc.…”
