2020) Effect of Moringa oleifera seed extract on antioxidant activity and sperm characteristics in cryopreserved ram semen, ABSTRACT Semen cryopreservation damages sperm due to oxidative stress. This study evaluated the antioxidant capacity of Moringa oleifera seed extract in cryopreserved ram semen and the impact of the extract on sperm characteristics. Semen from eight hair rams (four rams per sampling) was allocated into four groups, according to their treatment prior to cryopreservation: Control (no extract), 0.5 (M0.5), 5.0 (M5.0), and 10.0 (M10) mg/mL of M. oleifera extract. The antioxidant activity (ferric reducing antioxidant power, FRAP) and the spermatic characteristics (sperm viability; progressive motility; fast motility; slow motility; acrosome damage; membrane damage; and mitochondrial activity) were assessed post-thawing. Variables were evaluated with analysis of variance followed by Tukey test. While no significant differences were detected in acrosomal damage, mitochondrial activity, fast-, or slow motility, the antioxidant activity was higher (P < 0.05) in M0.5 and M5 treatments. Viability and progressive motility increased in the M0.5 group (P < 0.05), whereas sperm membrane damage was lower (P < 0.05) in the same treatment. In conclusion, supplementation of ram semen with M. oleifera seed extract enhances antioxidant activity, sperm membrane integrity, viability, and progressive motility after thawing. This suggests that M. oleifera extract could be used as an antioxidant to improve the outcome of semen cryopreservation. Highlights. It is widely known that semen cryopreservation induces sublethal damage to sperm, deteriorating spermatic characteristics, which is largely attributed to oxidative stress. . There are scarce studies regarding the use of plant extracts as a replacement for conventional antioxidants to conserve sperm viability in cryopreserved ram semen. . Addition of Moringa oleifera seed extract to a concentration of 0.5 and 5.0 mg/mL prior to ram semen freezing increased antioxidant activity after cryopreservation. . M. oleifera seed extract at 0.5 mg/mL decreased post-thawing damage to the sperm membrane, increasing both viability and progressive motility. . Ultrasound-assisted extraction (UAE) and microwave-assisted extraction (MAE) of bioactive compounds, peels from Opuntia engelmannii cultivar (cv.) Valencia were optimized by response surface methodology. Randomized extraction runs were performed for each of the technologies employed in order to build effective models with maximum (bioactive molecules content and yield) and minimum (antioxidant activity) responses. A 5-level, 4-factor central composite design was used to obtain target responses as a function of extraction time (t), solid to liquid ratio (S/L), methanol concentration (metOH), and temperature (T). Specific response optimization for each technology was analyzed, discussed, and general optimization from all the responses together was also gather. The optimum values for each factor were: t = 2.5 and 1.4 min, S/L = 5 and 5 g/L, metOH = 34.6 and 0% of methanol and T = 30 and 36.6 °C, achieving maximum responses of 201.6 and 132.9 mg of betalains/g, 13.9 and 8.0 mg of phenolic acids/g, 2.4 and 1.5 mg of flavonoids/g, 71.8% and 79.1% of extractable solid and IC50 values for the antioxidant activity of 2.9 and 3.6, for UAE and MAE, respectively. The present study suggested UAE as the best extraction system, in order to maximize recovery of bioactive compounds with a high antioxidant activity.

Polygonum hydropiperoides é uma das plantas utilizadas como medicinal e que apresenta atividades biológicas confirmadas na dependência da composição química. Desta forma, estudos sobre a toxicidade e interação desta planta com as células são necessários, sendo os espermatozoides uma opção inovadora para a avaliação. Assim, o objetivo deste estudo foi determinar a composição química do extrato hidroalcoólico de P. hydropiperoides (PH) e a toxicidade em células espermáticas. Para a determinação da composição química do extrato foi utilizada a análise cromatográfica líquida de ultra eficiência. A toxicidade foi avaliada através da análise computadorizada da motilidade espermática de sêmen suíno doméstico (Sus scrofa domesticus), enquanto as alterações celulares foram analisadas por citometria de fluxo. Foram identificados no extrato vegetal um total de 17 compostos fenólicos destacando-se o ácido quínico e quercetina. Os parâmetros avaliados não sofreram alterações significativas nas concentrações de 1,5 até 6,2 mg.mL-1 independente do tempo de exposição ao extrato, sendo que a redução dos parâmetros ocorreu de forma progressiva ao aumento das concentrações (6,2 mg.mL-1 ≤ 12 mg.mL-1 ≤ 50 mg.mL-1 ≤ 100 mg.mL-1). Através da citometria de fluxo pode-se observar que em nível de estrutura celular, não foram observadas alterações significativas. As alterações provocadas pelo extrato podem sugerir os possíveis mecanismos de ação a serem explorados para aplicação clínica. O extrato hidroalcoólico de P. hydropiperoides, abundante em ácido quínico e quercetina, acarretou apenas em alterações na motilidade espermática, a partir de 12 mg.mL-1.
