Background Hippocratea celastroides Kunth, commonly known as “cancerina”, is used in Mexican Traditional Medicine for the treatment of gastric and intestinal infections, systemic and skin inflammation, injuries and gastritis. The aim of this research was to assess the anti-Helicobacter pylori activities of hydro-ethanolic root-bark extracts from Hippocratea celastroides Kunth in naturally infected dogs, after testing their acute and subacute toxicities in mice.MethodsTo determine in vivo acute toxicity, a hydro-ethanolic extract was obtained and administered orally in female and male Balb-C mice, at doses ranging from 2000 to 5000 mg/kg. For the subacute study, a hydro-ethanolic extract was given to male and female Balb-C mice at doses ranging from 200 to 2000 mg/kg body weight. The animals were observed daily over a period of 42 days for signs of toxicity. In the pre-clinical anti-Helicobacter spp. assay, 60 dogs were included. Eighteen and 19 dogs for the experimental and control groups respectively, concluded the study. The experimental treatment consisted of H. celastroides hydro-ethanolic extract and the control treatment of amoxicillin-clarithromycin-omeprazole.ResultsOral LD50 (lethal dose 50) values for hydro-ethanolic extract were indeterminable at the highest tested doses. Under the subacute administration, neither mortality nor any sign of toxicity were observed when the hydro-ethanolic extract was administered. There were no significant alterations in biochemical parameters. The prevalence of Helicobacter spp. infection in dogs was 97.1 % for the experimental group and 100 % for the control group. Effectiveness was of 33.3 and 55 % in the experimental and control group respectively. The oral administration of H. celastroides was well-tolerated and safe.ConclusionThe root-bark of H. celastroides produced no signs of toxicity, and manifested pharmacological activity that indicated the possibility of an alternative treatment for H. pylori infection. Currently, with reappraisal of ozone therapy, it has been utilized worldwide in research and clinical field. Most of the studies investigating effects of ozone on blood parameters are conducted by directly ozonating the blood. Rectal insufflation is a simple, easy and inexpensive method of delivering ozone. Little is known how these gases affect some fundamental hemorheologic parameters when given by insufflation. We aimed to investigate the effects of colorectally insufflated oxygenozone on red blood cell rheology in rabbits. Rabbits were divided into Group 1 (control); Groups 2, 3 and 4 (oxygen rectally insufflated respectively for 15, 21 and 36 days); Groups 5, 6 and 7 (ozone rectally insufflated respectively for 15, 21 and 36 days). Erythrocyte deformability, aggregation and osmotic fragility were determined from blood samples at the end of each treatment period. Our study showed an improvement in deformability, a decrease in aggregation and an increase in fragility following a 15 day ozone treatment. With longer ozone application the changes in aggregation and fragility returned back to control levels, however its effect on deformability sustained. Therefore, more than two weeks ozone insufflation may induce adaptation to changes induced by ozone suggesting its systemic effects.

In this study we investigated the effects of antibiotic and intraperitoneal ozone administration on pro-inflammatory cytokine, antioxidant levels and tissue damage in the treatment of experimentally infectious peritonitis. Thirty-three adult male New Zealand White Rabbits were used. The study consisted of four groups including the non-treatment group (G1), antibiotic group (G2), ozone group (G3) and ozone + antibiotic group (G4). Trimethoprim sulfadimethylprymidine was administered at a dose of 50 mg/kg subcutaneously (s.c.) and amoxicillin sodium at a dose of 15 mg/kg intramuscularly (i.m.). Medical ozone was administered intraperitoneally (i.p.) at a concentration of 30 µg O3/ml and dose of 80 ml/kg. Once peritonitis was produced, blood samples were taken from the animals before treatment and at regular intervals following treatment. Blood samples were used for haemograms and to measure levels of antioxidant and oxidative enzymes and pro-inflammatory cytokine levels. Tissue samples were examined histopathologically. There was no statistically significant difference between groups with respect to levels of pro-inflammatory cytokines. Antioxidant enzymes were found to be higher in groups G2 and G3. The granulocyte and lymphocyte values in group G3 were determined to increase earlier than in the other groups. The peritonitis scores were similar in G1 and G3, which is higher compared to G2 and G4 groups. Minimal tissue damage was observed in the group G2. It was concluded that antibiotic use for preventing peritoneum damage in experimental acute peritonitis was more effective than ozone therapy alone.
