Protective effects of 6‐gingerol on 6‐hydroxydopamine‐induced apoptosis in PC12 cells through modulation of SAPK/JNK and survivin activation

Abstract: Due to many therapeutic effects, Ginger (Zingiber officinale) is the most widely used spice around the world, including in Iran. Due to its potent anti-inflammatory and antioxidant effects, ginger may protect against neurodegenerative disorders. Here, we investigated the effects of 6-gingerol (the main bioactive compound in ginger) on 6-hydroxydopamine (6-OHDA)-induced cell death in PC12 cells. Cell viability, cell apoptosis, and stress-activated protein kinase/c-Jun N-terminal kinase (SAPK/JNK), and survivin … Show more

“…A human proximal tubular epithelial cell line (HK-2) was used to determine the cytotoxicity of the peptides via the Resazurin cell viability kit. 58 Briefly, 1 × 10 4 cells/well were plated in 96-well plates (containing 10% serum) and incubated for 12 h; then, the supernatant was discarded. The cells were followed by treatment with various concentrations of peptides for 1 h (without serum) at 37 °C in 5% CO 2 .…”

“…37 Furthermore, a recent study demonstrated that 6gingerol significantly inhibited 6-OHDA-induced cell apoptosis of PC12 cells, which was accompanied by lower levels of the activated form of stress-activated protein kinase/c-Jun Nterminal kinase (SAPK/JNK). 38 Hence, zingerone, 6-shogaol, and 6-gingerol may be the main ginger components with antioxidant properties in PD.…”

“…In particular, 6-shogaol pretreatment has been associated with reduced lipopolysaccharide (LPS)-induced nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression in these cells . “…In addition, 6-gingerol has excellent blood-brain barrier permeability, resulting in direct effects on brain tissue ( Simon et al, 2020 ). Importantly, relevant studies have confirmed that 6-gingerol also exhibits neuroprotective effects against hypoxic-ischemic brain injury ( Zhao et al, 2021 ), cerebral ischemia/reperfusion injury ( Luo Y. et al, 2021 ), amnesia ( Kim et al, 2018 ), acrylonitrile-induced brain damage ( Farombi et al, 2018 ) and Alzheimer’s disease ( Rezazadeh-Shojaee et al, 2021 ). However, the protective effects of 6-gingerol against EBI post-SAH remain unclear.…”

“…6-gingerol is a natural compound extracted from ginger and has been reported to exhibit potent effects against oxidation, inflammation, apoptosis and cancer ( Chen et al, 2018 ; de Lima et al, 2018 ; Zhang et al, 2018 ; Luo J. et al, 2021 ; Rezazadeh-Shojaee et al, 2021 ). Previous studies have confirmed that 6-gingerol ameliorates apoptosis under different experimental conditions in various cells and organs, such as hypoxia-induced cardiomyocytes ( Zhang et al, 2019 ), myocardial ischemia/reperfusion injury ( Lv et al, 2021 ), myocardial fibrosis ( Han et al, 2020 ), osteoarthritis ( Abusarah et al, 2017 ), renal damage ( Hegazy et al, 2016 ) and atherosclerosis ( Yang et al, 2020 ).…”
