Protective Effect of Resveratrol against Pentylenetetrazole-Induced Seizures and Its Modulation by an Adenosinergic System

Abstract: The effect of trans-resveratrol (resveratrol), a polyphenolic compound with potent antioxidant activity, was investigated against pentylenetetrazole (PTZ) induced seizures in rats. Resveratrol (20, 40, and 80 mg/kg i.p.) administered 20 min prior to convulsive challenge with PTZ (60 mg/kg i.p.) dose dependently reduced the percent incidence of generalized tonic-clonic convulsions. Resveratrol (40 mg/kg) also potentiated the effect of sodium valproate (150 mg/kg) and diazepam (2 mg/kg) against PTZ-induced seizu… Show more

“…This compound, also known as 3,5,4 0 -trihydroxystilbene, is a naturallyoccurring phytoalexin that is found in multiple plant sources . In multiple models of neurological injury including stroke (Wang et al, 2002;West et al, 2007), spinal cord injury (Ates et al, 2006;Kaplan et al, 2005;Kiziltepe et al, 2004;Yang and Piao, 2003), epilepsy (Gupta et al, 2002a;2002b;Wu et al, 2009), and Huntington's disease (Parker et al, 2005), resveratrol has demonstrated efficacy in reducing neuropathological and behavioral sequelae. Some studies have also found that traumatic brain injury (TBI) in both adult (Ates et al, 2007) and immature (Sonmez et al, 2007) animals is amenable to treatment with resveratrol, although behavioral protection has not yet been demonstrated in adult models of TBI.…”

“…Recently, additional properties of resveratrol have been documented, such as radical scavenging, antioxidant activity 14,16 , neuroprotection 14,17 , antiviral activity 14,18 , antibacterial activity [19][20][21] , antitubercular activity [22][23][24] . Suitable substituted cisstilbene derivatives are characterized by potent inhibitory activity on COX-2, quite similarly with that observed for a variety of vicinal diarylheterocycles, among which important antiinflammatory drugs, like celecoxib and valdecoxib, are found 25 .…”
