Background/Aims: Lipopolysaccharide (LPS) induces acute lung injury (ALI) through oxidative stress and inflammation. Naringenin exerts antioxidant and anti-inflammatory effects. The possible protective effect of naringenin was investigated against ALI induced by LPS in rats. Methods: Rats received a single injection of LPS (5 mg/kg, i.v.). Naringenin was given for 4 consecutive days, at 2 doses (50 and 100 mg/kg/day, p.o.), starting 3 days before LPS administration. Results: LPS significantly increased wet/dry lung weight ratio, malondialdehyde, nitric oxide (NO), interleukin-6, and myeloperoxidase activity in the lung tissues. Naringenin, particularly the higher dose, significantly ameliorated the LPS-induced changes in the measured parameters. Also, naringenin markedly reduced the histopathological lung tissue injury that resulted from LPS. Naringenin significantly decreased the LPS-induced expression of nuclear factor-κB, inducible NO synthase, tumor necrosis factor-a, caspase-3, and significantly increased heat shock protein 70 expression in the lungs. Conclusion: Naringenin significantly protected against LPS-induced ALI in rats through its anti-inflammatory, antioxidant, antinitrosative, and antiapoptotic effects.

Aluminum chloride (AlCl3) and acrylamide (ACR) are well known as environmental pollutants inducing oxidative stress. Our study investigated the effects of these contaminants and if the hydrophilic fraction of extra virgin olive oil was able to prevent lung oxidative stress and DNA damage. Animals were divided into four groups of six each: group 1, serving as controls, received distilled water; group 2 received in drinking water aluminum chloride (50 mg/ kg body weight) and by gavage acrylamide (20 mg/kg body weight); group 3 received both aluminum and acrylamide in the same way and the same dose as group 2 and hydrophilic fraction from olive oil (OOHF) (1 ml) by gavage; group 4 received only OOHF by gavage. Exposure of rats to both aluminum and acrylamide provoked oxidative stress in lung tissue based on biochemical parameters and histopathological alterations. In fact, we have observed an increase in malondialdehyde (MDA), H2O2, and advanced oxidation protein product (AOPP) and a decrease in reduced glutathione (GSH), non-protein thiols (NPSH), and vitamin C levels. Activities of catalase (CAT), glutathione peroxidase (GPx), and superoxide dismutase (SOD) were also decreased. Histopathological changes in lung tissue were noted like emphysema, vascular congestion, and infiltration of inflammatory cells. A random DNA degradation was observed on agarose gel in the lung of AlCl3 and acrylamide (ACR)-treated rats. Co-administration of OOHF to treated rats improved biochemical parameters to near control values and lung histoarchitecture. The smear formation of genomic DNA was reduced. The hydrophilic fraction of extra virgin olive oil might provide a basis for developing a new dietary supplementation strategy in order to prevent lung tissue damage.

A B S T R A C TZataria multiflora Boiss (Z. multiflora) belongs to the Lamiaceae family is used traditionally for culinary and medicinal purposes. Different pharmacological effects have been described for the plant including; bronchodilation, vasodilation, and effect on lung inflammation. The plant is also used as a remedy against cough in the traditional medicine. In this article, 'pharmacological effects of Z. multiflora and its constituents focusing on their anti-inflammatory, antioxidant, and immunomodulatory properties were reviewed' by searching various databases until 'June' 2016. The anti-inflammatory effects of the plant such as decreased total white blood cell, neutrophils, and eosinophils counts were demonstrated. The sprotective effects of Z. multiflora on serum levels of phospholipase A2 and total protein were showed. In addition, constituents of the plant, such as flavonoids and carvacrol, also showed anti-inflammatory effects. Z. multiflora also reduced oxidative stress by scavenging free radicals and can be used in the therapy of oxidative damage. Decreased level of malondialdehyde and protective effects of Z. multiflora on serum levels of nitric oxide were also shown. Improvement of the serum levels of IgE, reduction in pro-inflammatory cytokine , and increased anti-inflammatory cytokines (IFN-c and FOXP3) were shown for Z. multiflora and its constituents, carvacrol and thymol. These results indicated that carvacrol and thymol could be used for treatment of inflammatory diseases as new anti-inflammatory agents. Therefore, Z. multiflora showed anti-inflammatory, antioxidant, and immunomodulatory effects which could be used for treatment of inflammatory and immune dysregulation diseases or disorders associated with increased oxidative stress.
