The aim of this study was to evaluate the effect of farnesol on the production of acids and hydrolytic enzymes by biofilms of and. The present study also evaluated the time-kill curve and the effect of farnesol on matrix composition and structure of single-species and dual-species biofilms. Farnesol, at subinhibitory concentrations, showed a significant reduction in biofilm acid production, but did not alter hydrolytic enzyme production. The number of cultivable cells of both microorganisms was significantly reduced after 8 h of contact with farnesol. Extracellular matrix protein content was reduced for biofilms formed in the presence of farnesol. In addition, confocal laser scanning and scanning electron microscopy displayed structural alterations in all biofilms treated with farnesol, which included reduction in viable cells and extracellular matrix. In conclusion, farnesol showed favorable properties controlling some virulence factors of and biofilms. These findings should stimulate further studies using this quorum-sensing molecule, combined with other drugs, to prevent or treat biofilm-associated oral diseases.

Rapamycin is an important natural macrolide antibiotic with antifungal, immunosuppressive and anticancer activity produced by Streptomyces hygroscopicus. In this study, a mutant strain obtained by ultraviolet mutagenesis displayed higher rapamycin production capacity compared to the wild-type S. hygroscopicus ATCC 29253. To gain insights into the mechanism of rapamycin overproduction, comparative metabolic profiling between the wild-type and mutant strain was performed. A total of 86 metabolites were identified by gas chromatography-mass spectrometry. Pattern recognition methods, including principal component analysis, partial least squares and partial least squares discriminant analysis, were employed to determine the key biomarkers. The results showed that 22 potential biomarkers were closely associated with the increase of rapamycin production and the tremendous metabolic difference was observed between the two strains. Furthermore, metabolic pathway analysis revealed that amino acids metabolism played an important role in the synthesis of rapamycin, especially lysine, valine, tryptophan, isoleucine, glutamate, arginine and ornithine. The inadequate supply of amino acids, or namely "nitrogen starvation" occurred in the mutant strain. Subsequently, the exogenous addition of amino acids into the fermentation medium of the mutant strain confirmed the above conclusion, and rapamycin production of the mutant strain increased to 426.7 mg/L after adding lysine, approximately 5.8-fold of that in the wild-type strain. Finally, the results of real-time PCR and enzyme activity assays demonstrated that dihydrodipicolinate synthase involved with lysine metabolism played vital role in the biosynthesis of rapamycin. These findings will provide a theoretical basis for further improving production of rapamycin.

Cloning, expression pattern, and potential role of MAPKp38 and NF-κBp65 in response to lipopolysaccharide in yellow catfish (Pelteobagrus fulvidraco)

Mitogen-activated protein kinases (MAPK) and nuclear factor kappa B (NF-κB) pathways are considered to be two crucial intracellular signaling cascades in pro-inflammatory responses. In this study, we reported the coding sequences (CDS) of MAPKp38 and NF-κBp65 from yellow catfish. We also investigated the gene structure, expression patterns and functional role in yellow catfish. The CDS of MAPKp38 is 1,086 bp encoding 361 amino acids (AA). The MAPKp38 protein has a long highly conserved serine/threonine protein kinases catalytic domain. The NF-κBp65 CDS is 1,794 bp, and the gene encodes 597 AA, with a Rel homology domain (RHD) which consists of a RHD-DNA-binding domain and an Ig-like, plexins, transcription factors (IPT) domain. Moreover, MAPKp38 and NF-κBp65 protein of bony fish and other vertebrates have a single clade. Quantitative real-time PCR analysis revealed the presence of the MAPKp38 and NF-κBp65 transcript in 12 tissues of healthy yellow catfish. The highest expression levels of MAPKp38 and NF-κBp65 were detected in the heart and liver, respectively. Upon stimulation with an intraperitoneal injection of lipopolysaccharide (LPS), the expression levels of MAPKp38 and NF-κBp65 were up-regulated in the intestine. These results indicated that MAPKp38 and NF-κBp65 play important roles in mediating the response protection against LPS in yellow catfish.
