beta-Microseminoprotein (MSP) is one of the most abundant proteins in human seminal plasma and is secreted from the prostate gland. Its evolution can be traced from primates down to nonvertebrate species such as amphioxus, despite substantial differences in the primary structure. Most mammals are known to have one single MSP gene, but we have previously shown that the cotton-top tamarin and the common marmoset-two New World monkeys-carry several MSP genes. In this study we continue our characterization of MSP genes in the cotton-top tamarin by presenting the full nucleotide sequence of the three previously identified genes, mspA, mspE, and mspJ. A promoter analysis using the luciferase reporter showed that mspE is as transcriptionally active as the single human MSP gene, whereas mspA and mspJ display no activity with this assay. Two novel MSP genes were also identified, mspB and mspH, both of which are pseudogenes. MspB has a frameshift mutation in the third exon resulting in a new C-terminus and premature stop of translation. MspH has the features of a processed pseudogene, originating from a transcript of mspE. It is integrated into the genome together with another processed pseudogene originating from a transcript of the nucleoporin gene NUP88. The MSP genes described in this study probably arose by phylogenetically rather late duplication or retrotransposition, suggesting that they are confined to a limited number of New World monkeys.

Fluorescence and NMR Studies of Human Seminal Plasma Prostatic Inhibin: Association of Lifetimes with Sterically Constrained Tryptophans

Human seminal plasma prostatic inhibin (HSPI) is one of the first seminal plasma proteins, which has been isolated, purified, and characterized. HSPI contains two tryptophans at positions 32 and 92 along its 94 amino acid primary sequence. Among the three fluorescence quenchers acrylamide (neutral), potassium iodide (anionic), and cesium chloride (cationic), potassium iodide is found to quench the tryptophan fluorescence more compared to the other two. The fluorescence decay of HSPI is biexponential with lifetimes 5.86 and 2.44 ns. The Stern−Volmer quenching curves in the case of fluorescence intensity and average lifetime are identical, indicating that the quenching mechanism is purely dynamic. The decay associated spectra of the two lifetimes show that the two tryptophans are solvent-exposed. The fluorescence quenching data is in favor of associating the two lifetimes to separate tryptophans. The fluorescence anisotropy decay of HSPI is single exponential with a correlation time of 9.2 ns, which is due to the rotation of entire protein. Absence of any fast component in the anisotropy decay indicates that the two tryptophans are in motionally restricted, rigid environments. In NMR studies, the cross-peak patterns observed in 2D-COSY and 2D-NOESY spectra of HSPI gave unambiguous evidence that each of the two tryptophans is sterically constrained and exists in a single rotamer population.

Effect of prostatic inhibin peptide (PIP) on prostate cancer cell growth in vitro and in vivo

Prostatic inhibin peptide (PIP), is a 94 amino acid protein which is secreted by the prostate gland in an androgen-independent manner. Previously, it has been demonstrated that PIP appears to inhibit follicle-stimulating-hormone (FSH) secretion by the pituitary and prostate glands. In vitro, the Dunning R3327 rat prostate cancer cell line MAT-LyLu (MLL) cells and the human prostate cancer cell line PC-3, are stimulated to grow in response to exogenous FSH and these effects are blocked by PIP. In vivo, PIP inhibits the growth of the highly metastatic MLL prostate cancer cell line. A comparison of hormone levels in control and PIP-treated rats demonstrates a significant inhibition of FSH in treated animals. It appears that, in vivo, PIP may inhibit prostate cancer growth by inhibiting FSH. PIP may represent a novel hormonal treatment for prostate cancer.
