Zusammenfassung Die Bewertung von Verbraucherexpositionen umschließt alle Quellen und Pfade,über die Personen im privaten und häuslichen Umfeld mit Chemikalien in Kontakt kommen.Dabei sind Verbraucher nicht nur diejenigen,die selbst Verbraucherprodukte anwenden,sondern auch alle weiteren Personen,die sekundär (indirekt) mit dem Produkt Kontakt haben,z.B.Familienangehörige.Eine Verbraucherexposition kann gemessen oder geschätzt werden.Beide Verfahren haben Vor-und Nachteile.Während sich Messungen meistens auf individuelle Expositionssituationen beziehen,beschreiben Modelle eher eine verallgemeinerte Expositionssituation (Szenario).Messungen haben daher ihren Platz, wo Quellen von Schadstoffen aufgespürt werden müssen,Modelle eher dort,wo systematisierte Betrachtungen von Risiken durchgeführt werden.Zur Expositionsschätzung gehören 3 Abschnitte,erstens die Charakterisierung der Expositionsquelle (z.B.eine Haushaltschemikalie) mit Beschreibung der Anwendungsmodalitäten (Häufigkeit,Dauer); zweitens die Beschreibung des zeitlichen Verlaufes der Konzentration des Stoffes im Kontaktmedium,vor allem in der Luft und auf der Haut.Die Exposition wird darüber hinaus durch das Verhalten der exponierten Person selbst bestimmt.Hier ist die Kontaktdauer, auch Zeitbudget genannt,von großer Bedeutung.Besondere Verhaltensmuster können auch für bestimmte Populationen von Bedeutung sein.Man nimmt an,dass z.B.im Kindesalter die orale Aufnahme durch das sog. Mouthing (Hand-in-den-Mund-Stecken) eine besondere Rolle spielt.Die Risikocharakterisierung erfolgt durch einen Vergleich der Exposition,ausgedrückt als Konzentration oder Dosis,mit dem NOAEL 1 oder LOAEL 2 .Je geringer der Unterschied zwischen beiden Werten ist,desto größer ist die Wahrscheinlichkeit des Eintretens eines gesundheitlichen Schadens. In diesem Beitrag werden nun die wesentlichen Grundprinzipien und Vorgehensweisen der Expositionsermittlung dargestellt. Schlüsselwörter Exposition · Verbraucher · Expositionsszenarien · Expositionsschätzung der Atemluft, auf der Haut oder in dem Material, das verschluckt werden kann. Die interne Exposition wird über die Schätzung der Menge ermittelt, die über die Lungen, die Haut oder über den Magen-Darm-Trakt resorbiert werden kann. Die Höhe der internen Exposition ist darüber hinaus von den Mechanismen der Verteilung des Stoffes im Kör-per und seiner Elimination (Metabolismus und Ausscheidung) abhängig. Diese Arbeit versucht, die wesentlichen Grundprinzipien und die damit verbundenen Schritte der Expositionsermittlung darzustellen und in ein Gesamtkonzept einzubinden, das sich in erster Linie an den Anforderungen der Modellierung ausrichtet. Glycol ethers still continue to be a workplace hazard due to their important use on an industrial scale. Currently, chronic occupational exposures to low levels of xenobiotics become increasingly relevant. Thus, sensitive analytical methods for detecting biomarkers of exposure are of interest in the field of occupational exposure assessment. 1-Methoxy-2-propanol (1M2P) is one of the dominant glycol ethers and the unmetabolized urinary fraction has been identified to be a good biological indicator of exposure. An existing analytical method including a solid-phase extraction and derivatization before GC/FID analysis is available but presents some disadvantages. We present here an alternative method for the determination of urinary 1M2P based on the headspace gas chromatography technique. We determined the 1M2P values by the direct headspace method for 47 samples that had previously been assayed by the solid-phase extraction and derivatization gas chromatography procedure. An inter-method comparison based on a Bland-Altman analysis showed that both techniques can be used interchangeably. The alternative method showed a tenfold lower limit of detection (0.1 mg/L) as well as good accuracy and precision which were determined by several urinary 1M2P analyses carried out on a series of urine samples obtained from a human volunteer study. The within- and between-run precisions were generally about 10%, which corresponds to the usual injection variability. We observed that the differences between the results obtained with both methods are not clinically relevant in comparison to the current biological exposure index of urinary 1M2P. Accordingly, the headspace gas chromatography technique turned out to be a more sensitive, accurate, and simple method for the determination of urinary 1M2P.
