Prophylactic role of coenzyme Q10 and Cynara scolymus L on doxorubicin-induced toxicity in rats: Biochemical and immunohistochemical study

Abstract: Objective:The study aims to evaluate the protective effects of coenzyme Q10 (CoQ10) and Cynara scolymus L (CS) on doxorubicin (dox)-induced toxicity.Materials and Methods:Sixty male rats were divided into six groups. Group 1 as a control. Group 2 received dox (10 mg/kg) intraperitoneally. Group 3 received CoQ10 (200 mg/kg). Group 4 received CS (500 mg/kg). Group 5 received CoQ10 (200 mg/kg) and dox (10 mg/kg). Group 6 received CS (500 mg/kg) and dox (10 mg/kg). The rats were then evaluated biochemically and im… Show more

“…GSH depletion leads to increased lipid Peroxidation and necrosis of hepatocytes. Furthermore, a significant increase in the MPO which removes H 2 O 2 and catalyzes the formation of toxic hypochlorous acid that leads to neutrophil infiltration and oxidative damage has been reported by Mustafa et al [13]. The present data is consistent with the previous study reporting that in the hepatic Caco-2 intestinal cells, AA induced oxidative stress through depletion of GSH, overproduction of ROS, and induction of apoptosis biomarkers that resulted in cell death [14].…”

“…Studies have shown that the CoQ10 has free radical-scavenging activities, liver injuryreducing properties and endothelial dysfunction improving [8]. It also has anti-oxidants, antifibrosis and anti-inflammatory effects [9].…”

“…IHC staining quantification was made using three non-overlapping fields per specimen, selected indiscriminately for analysis. Measurements were made with the use of NIS-Elements imaging software (Nikon, Brighton, MI) associated with an Nikon TE200U microscope and quantified using NIH ImageJ (v1.49) (http://rsb.info.nih.gov/ij) as previously described [20]. Briefly, the optical density (OD) of p-ALK immunostaining was evaluated with OD estimated using the following formula: OD = log (max intensity/mean intensity), where max intensity = 255 for 8-bit images [20].…”
