Impact of consuming a milk drink containing a probiotic on mood and cognition

Objective: The impact on mood and memory of consuming a probiotic containing milk drink, or a placebo, was examined as, previously, a poor mood has been found to correlate with the frequency of constipation. Design: A double-blind placebo-controlled trial with random allocation of subjects. Setting: Subjects went about their normal life in the community apart from three visits to the laboratory. Subjects: One hundred and thirty-two healthy members of general population, mean age 61.8 years, volunteered in response to local media coverage. One hundred and twenty-four finished the trial. Intervention: For a 3-week period, either a probiotic containing milk drink, or a placebo, were consumed daily. Mood and cognition were measured at baseline, and after 10 and 20 days of consumption. Results: At baseline those who reported themselves to be less frequently constipated were more clearheaded, confident and elated. Although the taking of the probiotic did not generally change the mood, this appeared to be a reflection of the generally good mood in this sample. When those in the bottom third of the depressed/elated dimension at baseline were considered, they selectively responded by reporting themselves as happy rather than depressed after taking the probiotic. The intervention did not, however, influence the reported frequency of defaecation, probably a reflection of the initially low incidence of constipation. An unexpected and possibly chance finding was that the consumption of probiotics resulted in a slightly-poorer performance on two measures of memory. Conclusions: The consumption of a probiotic-containing yoghurt improved the mood of those whose mood was initially poor. This improvement in mood was not, however, associated with an increased frequency of defaecation. Sponsorship: Funded by Yakult, Japan.

-Propionibacteria, which are largely used as Swiss-type cheese starters, also constitute promising human probiotics because of their beneficial effects on intestinal bifidobacteria and bacterial enzyme activities. To be efficient, they must survive acid stress imposed within the stomach and the surfactant effects of bile salts. We thus investigated the ability of Propionibacterium freudenreichii (strain Propiofidus ® SI41) to adapt to, and survive these stresses. In an in vivo study, 7 volunteers received Propiofidus ® SI41 in 3 different modes. The probiotic was supplied during 3 periods (i) in classical capsules at a low dose (5 × 10 9 CFU·d -1 ), (ii) in the same capsules at a higher dose (5 × 10 10 CFU·d -1 ), (iii) in acid-resistant capsules at the low dose. In the first case, propionibacteria were recovered above 10 5 CFU·g -1 in only 8 out of 14 faecal samples. In the two following cases, they were enumerated in all faecal samples between 10 5 and 10 7 CFU·g -1 . The recovery of live propionibacteria in faecal samples seemed to be related to a higher content in short chain fatty acids. This study, which clearly showed the role of gastric protection in the survival of propionibacteria, led us to investigate stress adaptation in this strain. Acid tolerance response was evidenced: transient exposure to pH 5 afforded protection towards acid challenge at pH 2. Moreover, we showed that pre-exposure to a moderate bile salts pre-treatment at 0.2 g·L -1 sharply increased its survival against a subsequent lethal challenge (1 g·L -1 ). Important changes in cellular protein synthesis were observed during adaptation using two-dimensional electrophoresis. Global protein synthesis decreased during adaptation treatments. In contrast, specific proteins were overexpressed during acid adaptation and others during bile salts adaptation. Among the up-regulated polypeptides, a carboxyl carrier protein (BCCP) and the recombinase RecR were identified during both stress responses while the universal chaperonins GroEL and GroES, as well as the replicase RepB, were only up-regulated during acid adaptation. These results bring new insights on the tolerance of P. freudenreichii to acid and bile salts, which should be taken into consideration for the development of probiotic preparations. INTRODUÇÃOCom o aumento na expectativa de vida da população, aliado ao crescimento exponencial dos custos médico-hospitalares, a sociedade necessita vencer novos desafios, através do desenvolvimento de novos conhecimentos científicos e de novas tecnologias que resultem em modificações importantes no estilo de vida das pessoas. A nutrição precisa se adaptar a esses novos desafios, através do desenvolvimento de novos conceitos. A nutrição otimizada é um desses novos conceitos, dirigida no sentido de maximizar as funções fisiológicas de cada indivíduo, de maneira a assegurar tanto o bem-estar quanto a saúde, como também o risco mínimo de desenvolvimento de doenças ao longo da vida. Nesse contexto, os alimentos funcionais e especialmente os probióticos e prebióticos são conceitos novos e estimulantes (Roberfroid, 2002).São considerados alimentos funcionais aqueles que, além de fornecerem a nutrição básica, promovem a saú-de. Esses alimentos possuem potencial para promover a saúde através de mecanismos não previstos através da nutrição convencional, devendo ser salientado que esse efeito restringe-se à promoção da saúde e não à cura de doenças (Sanders, 1998).
