These authors contributed equally to this work Aim: To investigate the predictive value of albumin (ALB) and γ-glutamyltransferase (GGT) in hepatocellular carcinoma (HCC) patients undergoing curative resection. We sought to establish a new scoring model for predicting the prognosis of HCC patients undergoing curative resection. Patients and methods: A retrospective analysis was performed in 303 HCC patients who underwent curative resection. Preoperative risk factors for survival were investigated using univariate and multivariate analyses. On the basis of significant factors, a prognostic scoring model was established. The overall survival (OS) and recurrence-free survival (RFS) were compared between different groups. Results: Multivariate Cox regression showed that preoperative decreased ALB levels and elevated GGT levels were significantly associated with poor OS and RFS. Multivariate analysis showed that ALB level, GGT level, portal vein tumor thrombus, and tumor number were independent prognostic factors for both OS and RFS. Thereafter, we established a preoperative prognostic scoring model combining the four risk factors. The results revealed that higher risk scores might mean worse OS and RFS. Conclusion: Preoperative ALB and GGT levels are potentially useful biomarkers for predicting the prognostic outcomes in HCC patients undergoing curative resection. Our new prognostic scoring model qualifies as a novel prognostic predictor for HCC patients after curative resection.

Aims Cachexia is defined as a complex metabolic syndrome that is associated with tissue damage. Some studies have shown that the liver metabolic alterations contribute to overall host tissue wasting. Knowing that leucine acts as cell signalling, we evaluated hepatic metabolism in Walker 256 tumour‐bearing rats and investigated the modulatory effects of a leucine‐rich diet. Methods and Results Wistar rats were distributed into 4 groups: control (C) and tumour‐bearing (W) groups, fed a control diet, and leucine (L) and leucine tumour‐bearing (LW) groups, which fed a leucine‐rich diet. After tumour evolution (21 days), liver samples were collected, and assessed the glycogen content via histological periodic acid‐Schiff (PAS) staining and performed the molecular and biochemical analysis. A higher liver‐to‐body weight rate was observed in W and LW groups, whereas a lower muscle‐to‐body weight ratio was observed only in W group. Hepatic glycogen content was lower only in W group, which had a greater number of hepatocyte nuclei; these parameters were unchanged in LW rats. Moreover, phosphoenolpyruvate carboxykinase (PEPCK), glycogen synthase, and lactate dehydrogenase (LDHA) gene expressions were higher in liver tissue from W group than in LW group. However, liver alkaline phosphatase and γGT activities, and also liver AMP‐activated protein kinase (AMPK) expression were higher in both tumour‐bearing groups. Conclusions Our results suggest that a leucine‐rich diet has a protective effect on the loss of skeletal muscle and also minimises the liver failure induced by Walker 256 tumours. Despite the lack of protection against liver damage, the leucine‐rich diet modulated liver energy stores, likely decreasing the futile Cori cycle and reducing energy expenditures.

Ginger and turmeric are commonly used in food as spices with wellreported antioxidant activity. However, hepatoprotective and nephroprotective effects of ginger and turmeric have not been adequately studied. Therefore, the current study used chicken model with acetone induced liver and kidney injuries to evaluate the hepatoprotective and nephroprotective activities of ginger and turmeric because metabolic functions of chicken and human liver is similar. Eighty cocks were randomized into 5 experimental groups of chicks. The first one served as control. While the second group was received 350mg acetone/kg body weight (given in drinking water) per day for 30 days. The third, fourth and fifth groups were received ginger, turmeric and mix of ginger and turmeric by 1% in food respectively after treatment with acetone. Hematological parameters, liver and renal function tests, hepatic oxidative stress enzymes, as well as, histopathological examination scores were determined. Results indicated that, administration of acetone induces alteration in various hematological parameters, hepatic enzymes and hepatic oxidative stress enzymes in chickens. Dietary supplementation of ginger (GEN) and turmeric (TUR) caused a significant amelioration in some hematological parameters in acetone treated chickens. GEN and TUR treatments could reduce damage induced by acetone in alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), gamma-glutamyl transpeptidase (GGT), glucose, total protein and their fractions levels. Furthermore, acetone promoted a significant increase in urea, uric acid and creatinine levels, while treatment with GEN or TUR or GEN+TUR at the same time of treatment with acetone maintained urea and uric acid concentrations at a level similar to the control group. Interestingly, GEN, TUR and GEN+ TUR treatments significantly ameliorated effects of acetone on chickens by improving the levels of the main antioxidant enzymes (superoxide dismutase SOD, glutathione-S-transferase GST) and malondialdehyde (MDA) a reliable marker of lipid peroxidation in the cells. Histologically, acetone appeared to be hepatotoxic in chick's. Hepatocytes were sensitive to the treatment with acetone and contained large number of congested blood vesicles, cytoplasmic vacuoles, pyknotic nuclei, and lymphocytic infiltration in treated animals. Turmeric and ginger treatments improved the vast majority of hepatic changes.
