Prognostic relevance of thymidine kinase isozymes in adult non- Hodgkin's lymphoma

Abstract: To determine whether kinase (TK) isozyme status adds clinically useful information in adult non-Hodgkin's lymphoma (NHL), we have analyzed peripheral blood plasma and lymphocytes of 44 patients with NHL for either TK1 or TK2 isozyme activity. On the basis of isozyme status, patients could be divided into two groups that did not differ significantly with respect to known determinants for survival. The median survival of patients exhibiting peripheral blood TK1 thymidine kinase activity was 40 wk and that of ind… Show more

“…With use of a recently developed enzyme assay, capable of detecting normal levels of S-TK, we have found elevated S-TK levels in patients with vitamin BIZ deficiency. These increased levels were not unexpected, as enhanced TK activity has been observed previously in patients with vitamin BIZ deficiency, both in the bone marrow (8) and in extracts of peripheral lymphocytes (9). To our knowledge, however, TK activity has not been investigated before in sera from patients with this disorder.…”

“…This is interesting in view of the findings of elevated S-TK levels in malignant disorders characterized by a rapidly proliferating unstable tissue, such as small carcinoma of the lung, non-Hodgkin's lymphoma of unfavourable histology and acute leukaemia (3,13). In addition, the degree of S-TK elevation in these conditions has been shown to be of prognostic value (3,9). A'question to be answered is whether the megaloblastic cells in vitamin BIZ deficiency contain higher levels of TK than normal or malignant blasts.…”

“…Another potential explanation considers leakage of TK1 and/or thymidine in the tumor microenvironment, which may also interfere with tumor [ 18 F]FLT uptake. 47,48 In addition, efflux pumps associated with chemotherapy resistance might also affect tumor [ 18 F]FLT uptake after treatment. 49 Finally, the lack of correlation might be unrelated to altered [ 18 F]FLT kinetics in response to treatment.…”

“…Future studies should include an assessment of survival following a standardised treatment protocol to assess the utility of sTK activity as a prognostic marker in feline lymphoma as it is in humans and dogs. 11,18 Such research should be guided towards investigating if cats with lymphoma and sTK values within the reference interval are more likely to achieve complete remission and longer survival than cats with lymphoma and high sTK activity. Furthermore, with increased case numbers it may be possible to compare sTK activity values from cats with lymphoma in different locations and of different phenotypes using immunohistochemistry.…”
