Profiling of circulating tumor cells in liquid biopsies from metastatic cancer patients

Abstract: Aim: Circulating tumor cells (CTCs) are crucial to tumor metastasis and valuable for prediction of clinical outcome in patients with solid tumors. Here, the authors aimed to establish a method for enumeration and characterization of CTCs from liquid biopsies. Methods: Peripheral blood mononuclear cells (PBMCs) were separated from blood samples from patients with metastatic cancer using Ficoll-Hypaque gradients and cultured to isolate and enumerate CTCs. Cultured CTCs were morphologically characterized by light… Show more

“…MMP-9 belongs to the metzincin family of mostly extracellularly operating proteases, and is a type IV collagenase and 92-kDa protein that is encoded by a gene located on chromosome 20 (20q13.12) in the human genome 44 . Studies over the last decades have proven the significant correlation between MMP-9 expression level in cells as well as extracellular vesicle exosomes and tumour metastasis 45 49 , which make it an important biomarker in liquid biopsy. A commercially available assay kit - SensoLyte ® 520 MMP-9 Assay Kit *Fluorimetric* (AnaSpec Inc.) was purchased and used to prepare the reaction mix for MMP-9 detection from both standard MMP-9 samples as well as MMP-9 samples collected from MDA-MB-231 cell culture supernatants.…”
