Production of IL-12 and IL-18 in human dendritic cells upon infection byListeria monocytogenes

Abstract: Dendritic cells (DCs) are major antigen-presenting cells of the immune system, which need to be activated in order to initiate an immune response. Here, we describe the immunostimulatory effects on human monocyte-derived DCs observed upon infection with Listeria monocytogenes or after treatment with listerial lipoteichoic acid (LTA) and lipopolysaccharide (LPS), respectively. All stimuli caused upregulation of costimulatory molecules, induced T-cell proliferative responses and secretion of cytokines in vitro. … Show more

“…LTA also induces less IL-12 than LPS and subsequently IFN-␥ (64,91). The cytokine pattern produced by LTA is similar to that induced by the whole bacterium (200,210).…”

“…This potential role of GM-CSF, in combination with other cytokines, for increasing the antibacterial competence of macrophages has already been advocated [22]. More specifically, GM-CSF could also cooperate for defense against Listeria infection with other proinflammatory cytokines, such as interleukin (IL)-12 [31,32] and IL-18 [33,34]. Intriguingly, the administration of a traditional Chinese medicine that protects against Listeria infection in mice has been shown to trigger the production of IL-1, IL-6, and GM-CSF [35].…”

“…Nevertheless, in a recent study in BALB/c mice, the cytokine gene expression of gastric tissue was functionally not impaired after L. monocytogenes infection (Park et al 2004). For IL-12 and IL-18, produced by L. monocytogenes-infected human DC (see: Kolb-Ma¨urer et al 2003a), gene expression was not reduced in the infected animals; on the contrary, it displayed a parallel increase with progressing (Park et al 2004), which might indicate a similar active synthesis in the mouse DC also.…”

“…Human DC are known to phagocytose L. monocytogenes and to subsequently mature (Paschen et al 2000;Kolb-Ma¨urer et al 2000, 2003a. It has been found that opsonization of L. monocytogenes with human serum significantly enhances phagocytosis of the DC via FccRIII (CD16)-mediated receptor uptake.…”
